Materials and methods: Swiss-albino mice, 3-4 months old, were used in this study. Taxol was administered by bolus intravenously through the tail vein. Radiation was given using a linear accelerator. There were four treatment categories, which comprised a total of 34 groups. Each group consisted of five animals. The first category was a control category which comprised one group (n = 5). The second treatment category was Taxol alone which comprised three groups (n = 15). The third treatment category was radiation alone which comprised three groups (n = 15). The fourth treatment category was Taxol plus radiation which comprised 27 groups (n = 135). Mice were killed 24 h after Taxol or radiation or combined administration using ether anesthesia. Using a light microscope, apoptotic and mitotic indices were counted on jejunal crypt cells of mice that were stained with hematoxylin-eosin. Differences between groups were statistically evaluated with Student's t-test. Results: Taxol caused a dose-dependent increase in apoptosis (P = 0.045) and decreased the mitotic index (P = 0.006) at high doses. Similarly, radiation caused a dose-dependent increase in apoptosis (P = 0.046) and decreased the mitotic index (P = 0.299) at higher radiation doses. Compared to radiation alone, Taxol caused a significant induction of apoptosis (P = 0.010). In combination, no significant radiosensitizing effect of Taxol was observed (enhancement ratio < 1), when compared to radiation alone. However, an increase in apoptosis was observed after 24 h of Taxol exposure when compared to 12 or 48 h of Taxol exposure (P = 0.0001 and P = 0.0001). Conclusion: These findings suggest that Taxol did not cause a radiosensitizing effect in intestinal crypt cells. However, a 24-hour pretreatment of Taxol exposure followed by radiation caused significant induction of apoptosis and reduction of the mitotic index when compared to other Taxol timing sequences. Thus, the lack of a radiosensitizing effect of Taxol in these proliferative cells may be due to enhanced mitotic death rather than apoptotic death.Publication Prevalence of transfusion transmitted virus infection and its effect on renal graft survival in renal transplant recipients(Informa Healthcare, 2002-01-01) Usta, M; Dilek, Kamil; Ersoy, A; Özdemir, B; Mıstık, R; Vuruşkan, H; Güllülü, M; Yavuz, M; Oktay, B; Yurtkuran, M; Usta, M; DİLEK, KAMİL; Ersoy, A; Özdemir, B; Mıstık, R; Mıstık, R; Vuruşkan, H; Güllülü, M; Yavuz, M; Oktay, B; Yurtkuran, M; Uludağ Üniversitesi/Tıp Fakültesi/Nefroloji Anabilim Dalı; Uludağ Üniversitesi/Tıp Fakültesi/Mikrobiyoloji Anabilim Dalı; Uludağ Üniversitesi/Tıp Fakültesi/Üroloji Anabilim Dalı; 0000-0002-7528-3557; X-8540-2019; AAH-5054-2021; AAZ-9075-2021; DFY-3761-2022; EFH-9523-2022; CTG-8811-2022; EHM-7377-2022; DKK-2716-2022; EGT-2006-2022Objective: Little is known about the prevalence of transfusion transmitted virus (TTV) infection in renal transplant recipients (RTxs) and its effects on allograft survival. We investigated the prevalence of TTV and its effects on liver injury and graft survival in RTxs.Material and Methods: The study was performed in 33 consecutive RTxs (8 females. 25 males) and 100 blood donors (35 females, 65 males). A nested polymerase chain reaction was used to detect TTV DNA in serum. Serum creatinine and alanine aminotransferase (ALT) levels and 24-h protein excretion were determined in both TTV-positive and -negative patients. The total number of blood transfusions, the duration of hemodialysis and the total duration after transplantation were recorded in RTxs. In addition, hepatitis B surface antigen (HbsAg), anti-hepatitis C virus (HCV) and hepatitis G virus DNA antibodies were determined in all patients. Results: TTV DNA was detected in 51.5% of RTxs and in 7% of the control group and this difference was statistically significant 3<0.01). In the RTx group, 64.7% of TTV-positive and 56.2% of TTV-negative patients had undergone a previous blood transfusion. However, the blood transfusion replacement rate, total duration of dialysis therapy and post-transplant period did not differ between these two groups. Five (15.1%) patients in the RTx group had abnormal liver function tests (ALT >40 IU/l). Of these patients, 2 were anti-HCV-positive, 1 was HBsAg-positive and anti-HCV- plus TTV DNA-positive and the serologic tests of the remaining 2 patients were all negative. Among the TTV-positive patients, 2 (11.7%) were anti-HCV-positive, 1 (5.8%) was HBsAg-positive and 3 (17.6%) were HGV DNA-positive. The baseline serum creatinine levels did not differ significantly between the TTV-positive and -neptive patients, being 1.5 +/- 0.6 and 1.4 +/- 0.6 mg/dl, respectively (p > 0.05). Two of the TTV-positive patients and 1 of the TTV-negative patients had proteinuria. A 1-year follow-up of TTV-positive and -negative patients demonstrated neither acute nor chronic graft rejection. Conclusion: In RTxs, TTV infection was more prevalent than in the normal population. In our patients the virus did not have an important effect on renal graft rejection and did not cause liver injury. However, the question of whether TTV infection may affect graft survival requires further long-term investigation in larger groups.Publication Investigation of genotoxic effect of taxol plus radiation on mice bone marrow cells(Wiley, 2002-01-01) Özkan, L; Egeli, Ü; Tunca, B; Aydemir, N; Çeçener, G; Akpınar, G; Ergül, E; Çimen, Ç; Özuysal, S; Kahraman-Çetintaş, S; Engin, K; Ahmed, M. Swiss-albino mice, 3-4 months old, were used in this study. Taxol was administered bolus intravenously through the tail vein. Radiation was given by using a linear accelerator. There were four treatment categories, which had a total of 34 groups. Each group consisted of five animals. The first was the control category that had one group (n=5). The second treatment category was taxol alone, which had three groups as per taxol dose alone (n=15). The third treatment category was radiation alone, which had three groups as per the radiation dose (n=15). The fourth treatment category was taxol plus radiation, which had 27 groups as per combined radiation dose plus taxol dose concentration and as per pre-treatment timing sequence of taxol before radiation (n=135). Mice were sacrificed 24 It after taxol or radiation or combined administration using ether anesthesia. The cells were then dropped on two labeled slides, flamed, air dried, and stained in 7% Giemsa; 20-30 well-spread mitotic metaphases were analyzed for each animal; the cells with chromosome breaks, acentric fragments, and rearrangements were evaluated on x 1,000 magnification with light microscope (Zeiss axioplan). The mitotic index was determined by counting the number of mitotic cells among 1,000 cells per animal. Differences between groups were evaluated with Student's t-test statistically, Taxol caused a dose-dependent increase in chromosomal aberrations (P=0.027). Similarly, radiation caused a dose-dependent increase in chromosomal aberrations (P=0.003) and decreased mitotic index (P=0.002). In combination, there were a small enhancements at the 40 mg/kg taxol dose level and at 0.25 and 0.5 Gy radiation doses in the 48 h group. However, an increase in chromosomal aberrations was observed after 48 hours of taxol exposure when compared 12 or 24 h of taxol exposure (P=0.001 and P=0.019). These findings suggest that taxol at the high doses with low dose radiation caused radiosensitizing effect in bone marrow cells. Forty-eight-hour pretreatment of taxol exposure followed by radiation caused significant induction of chromosomal aberrations and a reduction of mitotic index when compared to other taxol timing sequence. Optimization can be considered as one of the major quantitative tools in decision - making. Many problems are associated in the design, operation and productivity of textile mills under the force of increasing competition. These can be resolved only by applying optimization procedures. Optimization can take place at many levels and in many procedures in a textile company from the material supply to the delivery of the goods to the customer. Optimization problems can be found in management, in operations or in equipment A coordination of these levels is necessary for the technological management in the mill.Publication A research on controlled atmosphere (CA) storage of cv. A review of Turkish agricultural history shows that dramatic changes have occurred in fruit and vegetable industry. It seems evident that changes will continue for the coming years. Especially developments of South Marmara Regions fruit and vegetable processing industry provide a good example for the agri-food sector.Today, consumer preferences drive markets. Reshape of the international agricultural trade is required due to, mainly, the globalisation of the markets. This will give growers more competition and the issues of marketing, international trade, environmental protection and government policies will be important factors for them. The aim of this study it to examine the current situation of Turkish fruit and vegetable processing industry and highlight some points for the competitiveness of the industry. SWOT analysis was applied regarding the product quality, consumption, production, imports, exports and structure of market competition and distribution aspects.Publication Effect of some postharvest treatments and controlled atmosphere (ca) storage on basic quality criteria of peaches and nectarines(International Society Horticultural Science, 2003-01-01) Verlinden, B.E.; Nicolai, B.M.; DeBaerdemaeker, J.; Akbudak, B; Eriş, A; Bursa Uludağ Üniversitesi/Ziraat Fakültesi.; Verlinden, BE; Nicolai, BM; DeBaerdemaeker, JThe peach (Flavorcrest and Red Top) and nectarine (Fantasia and Fairlane) cultivars used in this study were stored in normal (NA) and controlled atmosphere (CA) conditions [(CO2%:O-2%) 3:3, 5:2, 10:2] consisting of 0+/-0.5degreesC temperature and 90+/-5% relative humidity in three years. Peaches and nectarines were subjected to some treatments (fungicide solution, low O-2 and intermittent warming treatment) either before or during storage. Physical and chemical changes (weight loss, respiration rate, fruit flesh firmness, titratable acidity, total sugar, overall appearance) were recorded at 15-day intervals during 45 day storage and after 10 day shelf life at the end of the storage. The results show that fruits of cv. Flavorcrest were stored more successfully in 5:2, fruits of cv. Red Top in 5:2 and partially 10:2, and fruits of cvs. Fantasia and Fairlane in 10:2 atmosphere combinations, compared with other treatments.Publication Determination of the crystallographic parameters of a fcc metal by means of electron channelling patterns(Wiley, 2003-01-01) Uğuz, Agah; UĞUZ, AGAH; Uludağ Üniversitesi/Mühendislik-Mimarlık Fakültesi; GFU-0210-2022Indexing and mapping of selected area electron channelling patterns (SAECPs) of an Al-Zn-Mg alloy have been performed. Although, channelling patterns are similar for crystals having the same system, since the interplanar spacing and the angular band width depend on lattice parameter, the details of the map will differ for every material. Angles between the poles and the angular separations of channelling bands were measured on the micrographs and calculated with the well known equations and found to be well in agreement within the error limits. Also the channelling line widths were calculated with the help of extinction distances of the reflecting planes. Structure factors and atomic scattering factors for selected reflections were also calculated for the present alloy whose a = 4.054 Angstrom.Publication Physical and biochemical changes during controlled atmosphere (CA) storage of cv. Granny Smith), two trials were carried out with the fruits taken from the same place during the successive two years. Following the harvest, fruits were transported to the cold storage and then they were stored in plastic cells of 120-L volume in which the inner atmosphere could be controlled. Having completed the precooling treatment, the fruit cv. Granny Smith were stored at 0+/-0.5degreesC and 90-95% relative humidity (RH) storage conditions under different CA combinations [(CO2%:O-2%) 0:21; normal atmosphere (NA)-control, 3:1, 3:1.5, 3:2 and 3:2.5]. After the storage period, they were kept at 20+/-2degreesC and 60-65% RH for 5 days so as to determine their shelf life. Some physical and biochemical analyses such as weight loss (%), respiration rate (Mg CO2/kg h), titratable acidity (TA) (%), pH, water soluble solids (WSS) (%), fruit flesh firmness (FFF) (lb), pectin esterase (PE) activity (ml/h), fruit skin colour (L, a, b) were conducted on samples which were taken at certain intervals during storage and shelf life periods. As a result, all CA applications were found better compared with the control application in cv. Granny Smith stored for 210+5 days. Among CA applications, 3:1.5 combination was determined to be better than the others.Publication Emotional stress and aging(Medimond Publishing Co, 2003-01-01) Park, AJ; Tokullugil, H. A; Bursa Uludağ Üniversitesi/Tıp Fakültesi/Biyokimya Anabilim Dalı.; Park, AJSenescence is that phase of the life span that is associated with an increasing probability of dying. A fundamental feature of senescence is "the diminished ability to maintain homeostasis".Stress is any real (physical.) or imagined (psychological and/or emotional) threatening situation, in which cellular hoincostasis is disturbed. In humans, mental or physical tension or strain, urgency, pressure, anxiety are conditions associated with stress. Physical or psychological stressors produce responses via the same cascade. Acute stress responses (the first wave) are immediate motor and cognitive responses. Chronic or prolonged stress responses (the second wave) are more delayed neuroendocrine responses. In humans, glucocorticoids (GCs); mainly cortisol increase to stress levels in the second wave, which is particularly responsible from the neuroendocrine effects.GCs have two types of receptors: Type I, mineralocorticoid receptor (MR, high affinity-low capacity) and Type II, glucocorticoid receptor (GR, low affinity-high capacity). MRs may play a role in regulating circadian changes in corticosteroid production. They regulate stress responses by negative feedback inhibition (NFBI) of the HPA-axis under physiological or basal conditions. Whereas, GRs become occupied during chronic stress and when there is excess release of GCs.The deleterious effects of GCs in the nervous system mainly are; disruption of learning, memory and plasticity (and cognition), inhibition of dentate neurogenesis, atrophy of the neuronal processes, endangerment of hippocampal neurones and neurotoxicity. GC exposure (i.e. basal concentrations) begin to increase somewhat linearly from late middle ages. "The feed-forward cascade" can partly explain the reason of this increase. Hippocampus is a negative feedback mediator of GC secretion. This is accomplished via hippocampal inhibitory projections to CRH neurones in the hypothalamus. Thus, hippocampal damage and down regulation of hippocampal GRs cause transient GC hypersecretion. Therefore, rising of basal levels of GCs and hippocampal neurone loss occur. So, impaired feedback mechanisms at the level of GRs contribute to "the hyperactivation of the HPA axis" with age. As a result clinically common findings of aging in humans at present can be summarised as following (note that, there are individual variations):1- Decline in cognitive vitality and learning and memory disorders2- Laboratory findings (without an existence of other medical comorbidities): a) Increase in the basal levels of HPA axis hormones: GCs (mainly cortisol), ACTH and CRH and b) Resistance to suppression tests.3- MRI (magnetic resonance imaging): Hippocampal atrophy and volume loss4- Histological findings (Practically not possible, only in biopsy specimens or in post-mortem analysis): Dendritic atrophy, Tau protein inclusions, and. neurone loss.So it can be concluded taht, "if we are sorry or make stress because of aging we must be aware of the danger that we may enhance the aging processes more".Publication Determination of some biochemical parameters in the seminal plasma of german shepherd dogs(Scientific Technical Research Council Turkey, 2003-01-01) Polat, Ü; Mustafa Kemal; SOYLU, MUSTAFA KEMAL; Kil, F.; Günay, G.; GÜNAY, ÜLGEN; Bursa Uludağ Üniversitesi/Veteriner Fakültesi/Dölerme ve Suni Tohumlama Anabilim Dalı.; Bursa Uludağ Üniversitesi/Veteriner Fakültesi/Biyokimya Anabilim Dalı.; ACB-3704-2022In this study, we aimed to determine the levels of some biochemical parameters in the seminal plasma of German Shepherd dogs.
