For this, 64 meat and 79 chicken Salmonella isolates, which were obtained from retail meat and farm samples analyzed in our laboratory from February 2006 to January 2009, were used. All Salmonella isolates were resistant to at least 4 antimicrobials tested, with the common highest resistance rates to erythromycin (100.0%), bacitracin (99.3%), and rifampicin (98.6%), regardless of the sample type. Additionally, meat isolates were 100.0% resistant to bacitracin, clindamycin, erythromycin, rifampicin, and chicken isolates were 100.0% resistant to erythromycin, spiramycin, penicillin G, doxycycline hydrochloride, and tetracycline. Four to 7 Multi Drug Resistant (MDR) Salmonella conformed 82.83% of all meat isolates, and 10, 12, 15, and 17 MDR Salmonella conformed 55.68% of all chicken isolates analyzed. The B/DA/E/RD and N/B/CT/E/SP/P/RD/DO/OT/TE resistances were the most common patterns as 35.94% and 13.92% within meat and chicken isolates, respectively. The considerably high resistance rates and MDR types to commonly used antimicrobial agents among the Salmonella isolates tested pose a significant risk factor for the treatment of foodborne Salmonella infections in humans.Item Assessment of microbiological changes in fresh uncooked Inegol meatballs stored under two different modified atmosphere packaging conditions(Ankara Üniversitesi, 2011) Temelli, Seran; Şen, Mehmet Kurtuluş Cem; Anar, Şahsene; Uludağ Üniversitesi/Veterinerlik Fakültesi/Gıda Hijyeni ve Teknolojisi Bölümü.; AAI-1092-2021; 6506404118; 36932470000; 6504131698This study was conducted to assess the microbiological changes in fresh uncooked Inegol meatballs stored at 4 degrees C under ambient (Control: C), and two modified atmosphere packaging (MAP) (M1: 30% O(2), 40% CO(2), 30% N(2) and M2: 60% O(2), 30% CO(2), 10% N(2)) conditions. pH measurements and microbiological analyses for total aerobic mesophilic bacteria (TAMB), Pseudomonas spp., Brochothrix thermosphacta (B. thermosphacta), Enterobacteriaceae, coliform bacteria, lactic acid bacteria (LAB), and yeast and molds were performed on days 0, 2, 5, 7, 10, 12, 15, 20 of storage. Packaging under ambient atmosphere resulted in an increase in the counts of all microorganisms and values. TAMB in C group composed of mainly Pseudomonas spp. and B. thermosphacta. There was restricted growth of Pseudomonas spp. in both MAP groups. There was no change in TAMB and B. thermosphacta counts of M1 group. in contrast to an increase in LAB counts. In M2 group, an increase in TAMB and a decrease in B. thermosphacta with no change in LAB counts was observed. Results indicate that M2 group MAP, with less growth of spoilage bacteria, was apparently more beneficial for refrigerated storage of Inegol meatballs.Item Assessment of microbiological contamination factors in frozen stuffed snail processing stages(Uludağ Üniversitesi, 2006-02-28) Günşen, Uğur; Dokuzlu, Uğur; Temelli, Seran; Şen, M. Cem; Uludağ Üniversitesi/Veteriner Fakültesi.In this study, six predetermined processing stages and six possible contamination sources in frozen stuffed snail production were examined for the enumeration of total aerobic mesophilic bacteria, coliforms, Escherichia coli (E. coli), coagulase positive staphylococci and for the detection of Listeria spp. and Salmonella spp. Results revealed that the cooling stage was a major contamination factor. Also, shells, although ‘pasteurized’, seemed to be one of the causes for the increases in the final microbial counts of the product. Another minor source for recontamination was determined as the cooling vat. Enteritidis ve S.Typhimurium varlığı yönünden tiplendirilmesi ve Gold Standart olan konvansiyonelserotiplendirmeye göre real time PCR’ın bu iki serovarın olası varlığının tespitindekietkinliğinin belirlenmesi amacı ile gerçekleştirilen çalışmada, 2021-2022 yıllarıarasında ISO 6579-1:2017 metodu ile izole edilmiş ve Salmonella spp. olduklarıdoğrulanmış, 104 adet karkas ve 57 adet yenilebilir iç organ kaynaklı olmak üzeretoplam 161 adet izolat ve aynı izolatlara ait PCR örnekleri analiz edildi. İzolatlarıntiplendirilmesinde konvansiyonel serotiplendirme ile S. Enteritidis ve S. Typhimuriumspesifik real time PCR (SE/ST-rPCR) analizi uygulandı.Çalışmada, tüm örnekler birlikte değerlendirildiğinde, konvansiyonelserotiplendirme ile izolatların %6,83’ü, SE/ST-rPCR analizi ile de %6,21’i serovarEnteritidis yönünden pozitif bulunurken her iki yöntemle de Typhimurium serovarınegatif olarak tespit edildi. Geleneksel serotiplendirme ile sırasıyla S. Virchow(%81,99), S. Schwarzengrund (%9,32), S. Enteritidis (%6,83), S. Bredeney (%0,62)serovarları bulunmuş olup izolatlardan ikisinin tiplendirilemediği belirlendi. SE/ST rPCR’ın relatif doğruluğu %99,37, duyarlılığı %90,91 ve özgünlüğü %100 ve her ikiyöntem arasındaki uyumun (κ: 0,94) neredeyse mükemmel olduğu saptandı.Sonuç olarak, yasal mevzuatta broyler örneklerinde aranması gereken ikiserovardan S. Typhimurium’un bulunmaması ve S. Enteritidis'in ise çok düşükprevalans oranlarında saptanmış olması, dominant serovarın S. Virchow ve ikinciprevalan serovarın ise S. Schwarzengrund olarak tespit edilmesi, broylerlerin S.Enteritidis ve S. Typhimurium dışında günümüzde farklı patojen serovarları dataşıdığını göstermesi yönünden güncel ve orijinal veri oluşturarak literatüre katkısağladı. Ayrıca, SE/ST-rPCR’ın Enteritidis ve Typhimurium serovarlarınınbelirlenmesinde konvansiyonel serotiplendirmeye alternatif olabileceği de belirlendi.Item Broyler kesim sürecinin farklı aşamalarından alınan örneklerde salmonella varlığı ve sayısının real time PCR ve ISO 6579-2:2012 ile belirlenmesi(Bursa Uludağ Üniversitesi, 2023-04-14) Demircioğlu, Ayşegül; Temelli, Seran; Bursa Uludağ Üniversitesi/Sağlık Bilimleri Enstitüsü/Veteriner Fakültesi/Besin Hijyeni ve Teknolojisi Anabilim Dalı.; 0000-0002-5121-2631Broyler kesim prosesinde soğutma öncesi (SÖ) ve soğutma sonrası (SS) karkas örneklerindeki Salmonella spp. varlığı ve sayısının belirlenmesi amacı ile 2021 yılında gerçekleştirilen çalışmada, Balıkesir’de faaliyet gösteren bir kanatlı kesimhanesinde kesilen toplam 480 broylere ait 96 adet birleştirilmiş boyun derisi örneği (48 SÖ ve 48 SS), ISO 6579-2:2012 Minyatürleştirilmiş En Muhtemel Sayı (mEMS) Tekniği kullanılarak Salmonella spp. varlığı ve seviyesi yönünden analiz edildi. İzolasyon sonrasında elde edilen izolatlar, Salmonella spp. spesifik r-PCR (Salm-PCR) analizi ile doğrulandı. Çalışmada incelenen SÖ örneklerin %97,92’sinin ve SS örneklerin %85,42’sinin toplam 96 örneğin ise 88’inin (%91,67) Salmonella spp. varlığı yönünden pozitif olduğu belirlendi. Örneklerdeki ortalama Salmonella sayısı ise SÖ’nde 1,84 log10 mEMS ve SS’nda 1,48 log10 mEMS değerlerinde bulundu. Hava soğutma sistemi kullanılarak yapılan soğutma işleminin patojenin sayısında 0,36 log10 mEMS’lık bir azalma oluşturduğu ve bu azalmanın da istatistiksel olarak önemli olduğu saptandı (p<0,05). Ülkemizde uluslararası referans metot ISO 6579-2:2012 ile bir kanatlı işletmesinde kesilen broyler karkaslarındaki Salmonella spp. yükünün ilk kez belirlendiği bu çalışmadan elde edilen sonuçlar, ilgili literatüre güncel ve orijinal veri katkısı sağladı. Ayrıca bu somut veriler, kanatlı işletmesinin son üründeki Salmonella yükünü azaltabilmesi için mevcut kesim prosesine yönelik yapılması gereken iyileştirmelerin planlamasında da yönlendirici bir rol oynadı.Item Bursa’da çiğ süt ve uht sütlerde aflatoksin m1 düzeyleri(Uludağ Üniversitesi, 2011-09-30) Oruç, H. Hüseyin; Temelli, Seran; Sorucu, Ali; Uludağ Üniversitesi/Veteriner FakültesiBu çalışmada, Bursa’da süt sığırcılığı işletmelerinden toplanan çiğ sütler ile perakende satışa sunulan UHT sütlerdeki aflatoksin M1 (AFM1) düzeylerinin belirlenmesi ve elde edilen sonuçların halk sağlığı yönünden değerlendirilmesi amaçlanmıştır. Bu amaçla, 30 adet çiğ süt ve 54 adet UHT süt olmak üzere toplam 84 adet süt örneğinde AFM1 düzeyleri ELISA tekniği ile belirlenmiştir. İncelenen çiğ süt ve UHT süt örneklerinin tümünde AFM1 tespit edilmiş olup (% 100.0), ortalama AFM1 miktarı 6.40±0.42 ng/kg olarak saptanmıştır. A total of 77 flocks (2153 tracheal swabs and blood samples) were sampled and all were tested by MG real time PCR (MG-rPCR) and MG-ELISA, and 32 flocks were tested by MS real time PCR (MS-rPCR). In the first part of this study covering 28 flocks, all samples from chickens with marked clinical symptoms and high MG-antibody levels gave negative results with MG-rPCR(1). Therefore, the MG-lipoprotein gene-specific primers (MG-rPCR(1)) of this PCR were replaced with MG-16S rRNA primers (MG-rPCR(2)), as were the MS-16S rRNA primers (MS-rPCR), thus the study was pursued accordingly. All of the first 28 flocks, which were 100% positive by MG-ELISA, were MG-rPCR(1) negative, whereas in the second part of the study, other 49 flocks, which were 87.8% MG seropositive, were found 42.9% positive by MG-rPCR(2). In addition, 5 selected flocks from the first 28 were negative, whereas 7.4% of the 27 selected flocks from the second 49 were positive by MS-rPCR. Overall, 81 out of 432 MG-rPCR(1-2) (18.7%) performed from 77 flocks, and 13 out of 187 MS-rPCRs (6.9%) in 32 flocks were determined as positive. ELISA results indicated that there could be significantly high false-positives in serological tests, thus results should not be relied upon one test system. One hundred dairy products (10 pasteurised milk, 10 kashar cheese, 20 white cheese, 18 mihalic cheese, 12 curd cheese, 20 ice cream, and 10 butter samples), collected randomly from food stores in Bursa, Turkey were analysed using the VIDAS LDUO and ISO 11290-1 methods. The results showed that one dairy sample (1%), a mihalic cheese, was positive for both Listeria spp. and L. monocytogenes. For this, 174 drag swab samples taken from 58 commercial layer flocks of 15, 25, and 40 week age were analyzed for Salmonella by ISO standard culture method (ISO 6579:2002/Amd 1:2007 - ISO) and by 2 real time polymerase chain reaction (rPCR) systems (Light Cycler SYBR Green I rPCR - LCSGIrPCR and DuPont BAX Q7 rPCR - BAXrPCR). Seventeen out of 58 (29.31%) flock samples, and 6 of the 15th and 25th weeks' (3.44% and 3.44%), and 10 (5.74%) of the 40th week's drag swab samples were Salmonella positive by all 3 methods. Twenty out of 22 Salmonella isolates (90.90%) were Salmonella enterica subsp. enterica, including serovars of 9 Enteritidis (SE) (40.9%), 7 Infantis (SI) (31.8%), 1 Hadar (4.5%), 1 Montevideo (4.5%), 1 Colombo (4.5%), 1 Spartel (4.5%), and 2 subspecies (9.10%) as Salmonella enterica subsp. arizonae and Salmonella enterica subsp. houtenae. Also isolates showed resistance to 23 of 24 antibiotics, with the highest resistances to Ampicillin (100%), Neomycin (100%), Penicillin G (100%) and Erythromycin (95.45%). Seventeen different control points and/or sample types (live snail, and snail meats after steaming, after shell removal, after first boiling, after gutting, after second boiling, after packaging and as frozen snail meat; air samples from gutting room, boiling room and packaging room; samples from gutting counter tops, package surfaces, scissors used during processing, forks used during processing, personnel hands, and potable water) have been examined for the enumeration of total aerobic mesophilic bacteria, coliforms, Escherichia coli, Enterobacteriaceae, coagulase positive staphylococci, Salmonella spp., Listeria spp., and yeast and molds. From the control points examined, raw material and environmental air were found as the primary contamination sources. Personnel hands and equipment used were determined as the secondary contamination sources. Second boiling and freezing stages during processing were determined to reduce the overall contamination rate, and therefore had positive effects on the microbiological quality of the final product. We determined: starter culture as the possible contamination source of coagulase positive staphylococci, enterococci and psychrophilic bacteria; brine and upper pressure plate as the contamination source for staphylococci; floor and packaging material as the contamination source of psychrophilic bacteria; cheese vat, cheese cloth and curd cutting knife as the contamination source of total aerobic mesophilic bacteria; cold room and production room air as the contamination sources for yeast and molds.Item Doğal koşullarda üretilen ve ısı işlemi uygulanan sucuklarda starter kültürlerin kullanım olanakları(Uludağ Üniversitesi, 1999-10-04) Anar, Şahsene; Soyutemiz, Ece; Temelli, Seran; Çetinkaya, Figen; Uludağ Üniversitesi/Veteriner Fakültesi/Besin Hijyeni ve Teknolojisi Anabilim Dalı.Bu çalışma starter kültür kullanarak doğal koşullarda üretilen sucuklara ısı işlemi uygulamasının sucuğun organoleptik, mikrobiyal ve kimyasal özelliklerine olan etkisinin incelenmesi ve olgunlaşma süresini kısaltmak amacı ile yapılmıştır. Çalışmada 3 grup sucuk üretildi. I. grup sucuklar doğal fermantasyon, II. grup sucuklar starter kültür kullanımı ve doğal fermantasyon işlemine tabi tutuldu. III grup sucuklara ise 20°C De 3 günlük olgunlaşmayı takiben merkezdeki ısı 63°C olacak şekilde 1 saat süre ile ısı işlemi uygulandı. I.ve II. grup sucuklar olgunlaşmanın 5. günü ve 18.günü mikrobiyolojik ve kimyasal analizlere tabi tutuldu. III grup sucuklar ise olgunlaşmanın I. günü, 3.gün, 30 dakikalık ısı işlemi sonrası ve 60 dakikalık ısı işlemi sonrası mikrobiyolojik ve kimyasal yönden incelendi. Results indicated that, compared to the control group, there was a significant and continuous decline in MG infection in chickens of Mentofin group determined by culture and Real-Time Polymerase Chain Reaction (MGrPCR) (P<0,05). Serology results in the control group indicated an increase in MG positivity from 25% to 40% (P>0,05), while there was no change in the Mentofin group (P>0,05). Culture results for MG positivity decreased from 85% to 5% in the Mentofin group, while this decrease was from 80% to 35% in the control group (P<0,05). There was a prominent decrease from 100% to 20% in MGrPCR positives in the Mentofin group (P<0,05) compared to a non-significant change observed from 95% to 80% in the control group (P>0,05). Results of this study indicate that Mentofin clearly had an effect on MG clearance from the tracheal epithelium, supported by detection of decline in MG infection in layers.Item Effect of rearing systems and shell color on some egg quality parameters(Uludağ Üniversitesi, 2016-12-12) Çetin, Ece; Temelli, Seran; Eyigör, Ayşegül; Uludağ Üniversitesi/Veteriner Fakültesi.; Uludağ Üniversitesi/Sağlık Bilimleri Enstitüsü.This study was conducted to determine some quality parameters of retail brown and white-shelled grade A table eggs, which were produced by cage, free-range and organic systems. For this, a total of 30 eggs, 10 from each rearing system, and 15 from each shell color were purchased, and egg weight, shape index, shell thickness, shell strength, and yolk color as outer and inner quality parameters were determined. The heaviest eggs were from cage system (65.04 g), and shape index and shell strength were the highest in organic eggs (78.37% and 41.02 N/cm2 ), while shell thickness was the lowest in free-range eggs (0.40 mm), and yolk color was the lightest in organic and free-range eggs (10 and 10.40). Additionally, brown eggs had higher values than white eggs in all of the parameters tested. Statistical analyses revealed that rearing systems have effect on shell thickness; while shell color has effect on both shell thickness and shape index (p<0.05). In conclusion, contrary to the consumer prejudice, retail eggs from different rearing systems do not exhibit significant differences on some quality parameters tested.Item Et parçalama ünitelerinde ve beyaz peynir üretiminde çalışan personel ellerinin hijyenik durumunun değerlendirilmesi(Uludağ Üniversitesi, 2006-02-27) Temelli, Seran; Şen, M. Cem; Anar, Şahsene; Uludağ Üniversitesi/Veteriner Fakültesi.Et parçalama üniteleri ve beyaz peynir üretiminde çalışan personel ellerinin hijyenik durumunun değerlendirilmesi amacıyla yapılan çalışmada, 2’şer adet kasap dükkanı ve hipermarket ile 2’şer adet mandıra ve süt fabrikası çalışanlarından üretim esnasında alınan toplam 80 adet örnek, toplam aerobik mezofilik bakteri, koliform bakteriler, Escherichia coli (E. coli), Enterobacteriaceae, stafilokoklar, koagülaz pozitif stafilokoklar ile maya ve küf sayıları yönünden incelendi. Koliform bakteriler, kasap dükkanları ve mandıralarda çalışan personelin ellerinde ortalama 103 kob/ml düzeyinde bulundu. E. coli’nin kasap çalışanlarında % 37.5, mandıra çalışanlarında ise % 28.5 oranında, hipermarket ve süt fabrikası çalışanlarında tespit edilebilir düzeyin altında olduğu ortaya konuldu. Kasap dükkanları ve hipermarketler ile mandıra ve süt fabrikalarında çalışan personel ellerinde stafilokokların sayısının sırasıyla ortalama 104 , 103 , 105 ve 104 kob/ml seviyelerinde bulunduğu; koagülaz pozitif stafilokokların ise kasap dükkanları ve mandıralarda çalışan personel ellerinde % 40 düzeyinde, süt fabrikalarında çalışan personel ellerinde ise % 5 düzeyinde olduğu tespit edildi. Çalışmanın sonucunda, özellikle kasap dükkanları ve mandıralarda çalışan personelin temel kişisel hijyen bilgilerinden yoksun olduğu; hipermarket ve süt fabrikalarında çalışan personelin ise almış oldukları hijyen eğitimleri sonucu öğrendiklerini uygulamada bir takım eksikliklerinin olduğu ortaya konuldu.Item Evaluation of ISO 6579 and FDA-BAM methods to complement real-time polymerase Chain reaction for the detection of salmonella in naturally contaminated poultry meat and red meat(Mary Ann Liebert, 2010-08) Eyigör, Ayşegül; Temelli, Seran; Çarlı, Kamil Tayfun; Uludağ Üniversitesi/Veterinerlik Fakültesi/Gıda Hijyeni ve Teknolojisi Bölümü.; Uludağ Üniversitesi/Veterinerlik Fakültesi/Klinik Öncesi Bilimler Bölümü.; AAI-1101-2021; AAI-1092-2021; E-3867-2010; 6602558950; 6506404118; 6601971539In this study, we evaluated the Salmonella detection capability and compatibility of a LightCycler polymerase chain reaction (LC PCR) system with two bacteriological methods, United States Food and Drug Administration's Bacteriological Analytical Manual Chapter 5: Salmonella (FDA) and International Organization for Standardization Method 6579 (ISO). The aim was to determine which bacteriological method would support LC PCR for testing naturally contaminated poultry and red meat samples with Salmonella. Twenty three (50.0%) and 24 (52.2%) out of 46 chicken meat samples were positive for Salmonella by the FDA and ISO methods, respectively. Five of the 15 (33.3%) turkey meat samples were found to harbor Salmonella by both bacteriological methods. None of the red meat samples were positive for Salmonella using the FDA method. There was one red meat sample (3.3%) positive for Salmonella using ISO method. LC PCR results indicated that 23 (50.0%) and 31 (67.4%) of the DNA templates obtained from the 46 preenriched chicken meat FDA and ISO samples were positive for Salmonella. Salmonella detection rate from turkey meat samples by ISO LC PCR was 6.7%, whereas no detection was observed by FDA LC PCR. FDA LC PCR detection rate in red meat samples was 23.3%, whereas the ISO LC PCR was 43.3%. Relative accuracy rates of ISO LC PCR and FDA LC PCR were 67.4%, 60.0%, 53.3% and 56.5%, 66.7%, 76.7% for chicken, turkey, and red meats, respectively. We presume that the low relative accuracy problem, which can be related to the use of FDA and ISO preenrichments for template preparations in the PCRs, can be overcome by the use of primary enrichments of both FDA and ISO bacteriologies.Item Geleneksel yöntemlerle üretilen kurutulmuş et ürünleri(Uludağ Üniversitesi, 2011-09-30) Temelli, Seran; Uludağ Üniversitesi/Veteriner Fakültesi.Geleneksel olarak üretilen kurutulmuş et ürünleri, Orta Doğu ve Güney Asya ülkeleri başta olmak üzere birçok ülkede halen tüketilmektedir.
