Item Akut ve kronik böbrek yetmezliğine sahip köpeklerde lipid profilinin değerlendirilmesi(Uludağ Üniversitesi, 2003-04-24) Şentürk, Sezgin; Çetin, Meltem; Gölcü, Esin; Udum, Duygu; Bursa Uludağ Üniversitesi/Veteriner Fakültesi.İnsanlarda serum lipid ve lipoprotein konsantrasyonları böbrek yetmezliği ile ilişkilendirilmiştir. Bu çalışmada total kolesterol, trigliserid, HDL-C, LDL-C’yi kapsayan serum lipidleri ile böbrek yetmezlikleri (akut ve kronik renal yetmezlik) arasındaki ilişkinin araştırılması amaçlanmıştır. Kronik böbrek yetmezliğine sahip olan köpeklerin akut renal yetmezlikli ve sağlıklı köpeklerle karşılaştırılmasında, serum total kolesterol, trigliserid ve LDL-C seviyelerinin istatiksel olarak daha yüksek düzeyde olduğu bulundu (p<0.001). Bununla birlikte serum HDL-C seviyesinin kronik böbrek yetmezliği bulunan köpeklerde akut böbrek yetmezlikli ve sağlıklı köpeklere göre daha düşük düzeyde olduğu belirlendi (p<0.001). Material and Methods: Thirty primiparous Holstein dairy cows were selected for the study. Blood samples were collected from each cow seven days before the expected calving date, on the calving day, and 7, 14, and 21 days after calving. Blood non-esterified fatty acids (NEFA), beta-hydroxybutyric acid (BHBA), glucose, insulin, adiponectin, and TNF- alpha levels were measured. Revised Quantitative Insulin Sensitivity Check Index (rQUICKI) was calculated using data on NEFA, insulin, and glucose concentrations. Results: When compared to prepartum levels, serum concentration of adiponectin significantly increased on day 21 postpartum. The rQUICKI increased and NEFA levels decreased on day 7 after parturition. Insulin and glucose levels decreased on days 7, 14, and 21 postpartum when compared with prepartum levels. BHBA levels decreased on day 21 and TNF- alpha concentration also decreased on days 7, 14, and 21 postpartum. Adiponectin levels positively correlated with NEFA during the preparturient period. Negative correlation was detected between adiponectin and rQUICKI on calving day and on 14th day after parturition. TNF- alpha concentration positively correlated with glucose levels on day 7 prepartum and on 21st day postpartum and with rQUICKI on 21st day postpartum. Negative correlation was detected between adiponectin level and insulin sensitivity. Moreover, the most of the PNX-expressing neurons in the hypothalamus also co-express nesfatin-1. These outcomes may suggest that there is an interaction between PNX and nesfatin-1, at least in terms of neuroendocrine-mediated regulations. Hence, the study was planned to find out the effects of centrally delivered PNX and nesfatin-1 on male sex hormones or to show the interactive association of intracerebroventricularly (ICV) injected PNX+nesfatin-1 combination on the release of male hormones. PNX and nesfatin-1, single or together, were delivered ICV to different male Wistar Albino rat groups. Both PNX and nesfatin-1 induced a significant enhancement in plasma FSH, LH and testosterone without inducing any alteration in plasma GnRH in the rats. The central combinatorial treatment of both the neuropeptides produced a more potent rise in male plasma hormone levels than treating with single neuropeptide. In summary, our preliminary data show that centrally delivered PNX and nesfatin-1 can affect plasma male hormone levels. Total of 70 calves were divided into E. coli, C. parvum, C. parvum + viral, viral and control groups. Clinical parameters were recorded on days 0 and 7, post treatment. On these days, blood and stool samples were also taken. SAA, SCalp and FCalp levels were measured with commercial ELISA kits. Mean SAA levels on day 0 were measured as 0.54 (0.16-2.18) ng/ml and 38.40 (8.28-83.96) ng/ml for the diarrhea and healthy group, respectively (P < 0.001). Statistically significant difference was also obtained between the diarrhea [68.02 ng/ml 46.66-101.67] and control [24.05 ng/ml 11.92-56] groups in terms of SCalp levels on day 0 (P < 0.001). Cut-off value of SCalp to distinguish E. coli originated diarrhea from viral diarrhea was found to be 70.969 ng/ml (Sens: 94%). FCalp levels on day 0 were 93.08 (22.17-122.88) ng/ml and 87.01 +/- 3.33 ng/ml in the diarrhea and healthy groups, respectively (P = 0.04). Cut-off concentration of FCalp was found to be 91.804 ng/ml (P = 0.0057). In addition, as a result of the logistic regression analysis, FCalp's ability to identify animals with diarrhea was found to be 6.316 times (P = 0.009) higher. The highest levels of SCalp and FCalp measured on day 0 were found among E. coli group. Twenty four multiparous Holstein cows were sorted by parity, body condition score (BCS) in close-up period and season of calving and assigned to one of the three treatments. The cows in treatment 1 (T1) received two drenches at calving and 24h after calving. The cows in treatment 2 (T2) received three drenches at calving, 24h after calving and 7 days after calving. The cows in treatment 3 (T3) were the control. Each drench contained 143g of calcium as calcium propionate (0.68kg). Parameters studied were serum calcium, glucose and nonesterified fatty acid (NEFA) concentrations, dry matter intake (DMI), BCS, milk production (MP), incidence of retained placenta (RP) and metritis. Milk fever developed in 5 of 8 cows, in 3 of 8 cows and in 3 of 8 cows in T1, T2 and T3, respectively, at calving. There was no cow with milk fever in T1 and T2 at 4h after second drench (about 28h after calving) but 3 of 8 cows in T3 had still milk fever at this time. The cows receiving two drenches recovered from milk fever in a shorter term as compared to the cows in T3. There were no differences among treatments for DMI, BCS, MP, RP, serum glucose and NEFA concentrations during the experimental period. There was no difference for metritis between T1 and T3 but incidence of metritis in T2 was significantly lower as compared to T3 (P<0.05). Thirty-six male Saanen kids (36 +/- 14days of age) were used in the study. Each group consisted of 9 kids. The control group (G1) was fed with a diet that consisted of the above concentrated feed and oat hay, whereas the experimental groups consumed the same diet but with the concentrated feed uniformly sprayed with juniper essential oil 0.4 ml/kg (G2), 0.8 ml/kg (G3) or 2 ml/kg (G4). There were no differences (p>0.05) in live weight, live weight gain or feed consumption between the control and experimental groups. There was a significant improvement (p<0.05) in feed efficiency in the G3 group. There were no differences in the rumen pH, rumen volatile fatty acid (VFA) profile or faecal pH of the control and experimental groups. The rumen NH3N values were similar at the middle and end of the experiment, but at the start of the experiment, the rumen NH3N values differed between the control and experimental groups (p<0.05). The faecal score value was significantly (p<0.05) decreased in the experimental groups. The addition of juniper essential oil supplementation to the rations caused significant effects on the kids' antioxidant blood parameters. Although the superoxide dismutase (SOD) activity, total antioxidant capacity (TAC) and catalase values were significantly (p<0.05) increased in the experimental groups (G2, G3 and G4), especially group G4, the blood glutathione peroxidase (GPX) value significantly decreased in the experimental groups. Thirty healthy sheep subjected to analyze for phenotypic and gene Frequencies of K-c, Na-c and GSH concentrations. Correlation between these parameters and production/reproduction traits such as greasy fleece weight, milk production, birth rate. single and twin birth late and body weight were calcualated. A positive correlation was found between birth rate and mean K-c concentration (r = 0.481, P<0.008) and Na-c concentration (1-0.454. P<0.013). Another correlation between GSH concentration and greasy fleece weight (r=-0.368, P<0.049) and milk production (r=0.379, P<0.050) parameters axis also evidenced. The potassium and sodium concentrations were determined by flame photometry. The level of GSH in erythrocytes was determined by the DTNB method. The frequencies of the mentioned loci have been calculated and compared with some of the goat breeds studied so far by other authors. Our results indicated that there were bimodal distributions in K-e and GSH concentrations in Saanen goat breeds, whereas there was only GSH polymorphism in Turkish Hair Goats. The distribution of K-e concentrations Suggested the presence of high-K/low-K (HK/LK) polymorphism, based on the predetermined dividing value of 50 meq/L of K-e concentration in erythrocytes of Saanen Goats. Both types are shown to be controlled genetically by an autosomal locus with two alleles HK and LK, with HK being dominant over LK. While majority of the Turkish Hair Goat population (93%) had low (GSH(h)) or 'deficient' erythrocyte GSH concentrations (<50 mg/100 mL erythrocyte), Saanen breeds (93%) had high (GSH H) erythrocyte GSH levels. Relationships between the parameters were also evaluated. Forty lambs were used in a 49-day experiment. The lambs were group-fed either a basal diet (alfalfa hay and concentrate, n = 20) or the basal diet along with 5 g/day microalgae (n = 20). Feed intakes were recorded daily and body weight (BW) was measured weekly. Overall, microalgae feeding increased (P < 0.05) the BW and average daily gain. There were no significant differences for average feed intake or feed efficiency (P > 0.05). Microalgae feeding decreased rumen pH (P < 0.05) and tended to reduce proportion of acetate (P < 0.1), while it increased total rumen volatile fatty acid concentration (8.6%; P < 0.01) and proportions of propionate (13.9%, P < 0.01) and valerate (P < 0.01; 26.5%) compared with the control animals. Microalgae feeding increased (P < 0.05) blood glucose (98.47 vs. 84.97 mg/dL) and insulin (64.14 vs. 29.26 ng/mL), whereas it lowered total cholesterol concentrations in blood (62 vs. 58 mg/dL, P < 0.1) compared with the control animals. The results of this study indicate that microalgae supplement influences productivity and enhances dietary energy utilization in lambs during the summer.Item Influence of various volatile oils as a dietary supplement on biochemical and performance parameters in broilers(Uludağ Üniversitesi, 2015-04-28) Belenli, Deniz; Udum, Duygu; Cengiz, S. Şule; Polat, Ümit; Uludağ Üniversitesi/Veteriner Fakültesi.Volatile oils like rosemary, thyme and fennel oils have positive effects on poultry health and yields. The aim of this study were to investigate the effects of volatile oils on some serum biochemical parameters, growth hormone activities and growth performance in broiler chickens. A total of 180 broilers were randomly divided into 6 groups and 3 replicates of 10 animals according to supplementation of thyme oil, rosemary oil, fennel oil, oil mixture 100 ppm dosage and oil mixture 200 ppm dosage. Serum growth hormone activity and glucose concentration were not affected by dietary addition of volatile oils according to groups. However, addition of rosemary oil, thyme oil and fennel oil showed an effect on decreased serum cholesterol and partially lipid concentrations in the present study. In conclusion, supplementation of volatile oils had impressive effects on serum total cholesterol and total lipids levels in broiler diets. It is well known that nesfatin-1 collaborates with other central neuromodulatory systems, such as central corticotropin-releasing hormone, melanocortin, oxytocin, and cholinergic systems to show its autonomic effects. Central arachidonic acid cascade plays an important role to provide the homeostasis by exhibiting similar autonomic effects to nesfatin-1. Based on these similarities, the current study was designed to show the effects of intracerebroventricularly (ICV) injected nesfatin-1 on the hypothalamic arachidonic acid (AA) cascade.Immunochemistry and western blot approaches demonstrated that ICV administration of nesfatin-1 provokes an increase in the hypothalamic cyclooxygenase (COX) -1, -2 and lipoxygenase (LOX) protein expression. Moreover, the microdialysis study demonstrated that centrally injected nesfatin-1 increased the posterior hypothalamic extracellular AA products.In conclusion, these findings report that while nesfatin-1 is generating its autonomic effects, it also might be using central prostaglandins and leukotrienes by activating central COX and LOX pathways.Item Kıvırcık ırkı koyunlara ait bazı biyokimyasal değerlerin belirlenmesi üzerine bir araştırma(Uludağ Üniversitesi, 2014-03-26) Udum, Duygu; Üstüner, Hakan; Belenli, Deniz; Uzabacı, Ender; Uludağ Üniversitesi/Veteriner Fakültesi.Bu çalışmanın amacı Kıvırcık ırkı koyunlara ait kan örneklerinde bazı biyokimyasal parametrelerin belirlenmesidir. Araştırma, Manisa ili ilçelerinde yetiştirilen 5 farklı çiftlikten oluşan, 1-6 yaş aralığında, klinik olarak sağlıklı, 30 adet Kıvırcık ırkı koyunlarında yapıldı. Bu hayvanlara ait plazma ve serum örneklerinde, total kolesterol, total protein, trigliserit, fosfolipit, kalsiyum, inorganik fosfor ve glikoz, ticari spektrofotometrik kitler kullanılarak spektrofotometre yardımıyla belirlendi. Kıvırcık ırkı koyunlar arasında çiftliklere göre istatistiksel farkın anlamlı olmadığı saptandı (p<0.05). It has no inhibitory effects on acute/chronic food intake, and it has been reported by several researchers. The role of obestatin in metabolism is still not clear. In the present study, the purpose is to determine the effects of chronically administrated obestatin. For this purpose, (1 mu mol/kg; i. p.) or ghrelin (1 mu mol/kg; i. p.) and food restriction (24h fast: 24h fed) on plasma obestatin, ghrelin, leptin, insulin, cholecystokinin (CCK) and glucose levels, and body weight gain were investigated for 14 days in mice. Additionally, mice were treated with acute ip (100 nmol/kg) injections of obestatin or ghrelin to investigate the food consumptions, plasma obestatin and ghrelin levels to determine unknown acute effects of obestatin. Plasma ghrelin levels increased significantly in obestatin administered mice when compared with the control group for chronic treatment. This increase is consistent with immunohistochemical findings which claim that the number of ghrelin and obestatin immunopositive cells in fundus tissue of stomach are considerably high in obestatin treated animals. Plasma obestatin and ghrelin levels has shown an increase endogenously in food restricted mice, but plasma leptin and insulin levels have been found to be lower compared to the control group. Acute administration of obestatin caused a decrease in plasma obestatin level at 60 min after injection and had no effect on the reduction of food intake in each treatment time. To test the hypothesis that long term effect of peripheral ghrelin on serotonin and insulin levels in ruminant species, ten male lambs were 2 months old with an average body weight of 26 kg and 2 groups according to ghrelin. treatment in group I, animals were fed ad libitum in the group II, animals were fed ad libitum and intravenously injected with the ghrelin. In the laboratory, researchers have shown that ghrelin administration was significantly decreased plasma serotonin, insulin and glucose concentrations in the long term. Although, the results obtained by ghrelin. treatment in the long term are not enough clear in ruminant species and further research should validate the obtained results before applying this information on relationships between ghrelin, insulin and serotonin.Item Serum tumor necrosis factor - alpha alterations of dry cows vaccinated with inactive coronavirus, rotavirus, E. coli combined vaccine(Uludağ Üniversitesi, 2017-04-05) Mecitoğlu, Zafer; Akgül, Gülşah Demir; Catik, Serkan; Udum, Duygu; Batmaz, Hasan; Uludağ Üniversitesi/Veteriner Fakültesi/İç Hastalıkları Anabilim Dalı; Uludağ Üniversitesi/Veteriner Fakültesi/Biyokimya Anabilim Dalı.Tumor Necrosis Factor alpha (TNF-α) is a proinflamatory cytokine, closely associated with insulin resistance and related diseases such as type II ketosis and fatty liver of cows. Aim of the presented study was to investigate circulating levels of TNF-α after vaccinating dry cattle with inactive rotavirus, coronavirus and E.coli combined vaccine. 14 cattle were randomly assigned to two groups; vaccination group was vaccinated on days 0 and 21 of the study and control group not. Blood samples were collected on days 0, 1, 5, 15, 21, 22, 26, 36 of the study for evaluation of TNF-α and total leukocyte counts. Although not significant, TNF-α levels decreased in vaccination group after the second application. Total Leukocyte counts did not differ between and within groups. Results of the study demonstrates that vaccinating cattle with inactive rotavirus, coronavirus and E.coli combined vaccine in dry period does not induce alterations in circulating TNF-α levels.
