Item Arı sütünün ovaryum kanser hücrelerinin proliferasyonu ve apoptozisi üzerine etkisi(Bursa Uludağ Üniversitesi, 2022-06-16) Asmaz, Ender Deniz; Zik, Berrin; Bursa Uludağ Üniversitesi/Sağlık Bilimleri Enstitüsü/Veteriner Fakültesi/Histoloji ve Embriyoloji Anabilim Dalı.; 0000-0001-6468-8535Arı sütü, antitümörik, antibakterial, antiallerjik, antikanserojenik ve immünomodülatör gibi pek çok biyolojik aktiviteye sahip doğal bir birleşiktir. Ovaryum kanseri, en sık görülen kanser türleri arasında dördüncü sırada yer almakta ve özellikle epitelyal over kanseri, tüm ovaryum malign hastalıkların yaklaşık % 90'ını oluşturmaktadır. Sunulan çalışmanın amacı, arı sütünün farklı doz ve sürelerinin, epitelyal ovaryum kanserleri içinde görülme sıklığı en fazla olan seröz tipte epitelyal over kanseri üzerine proliferatif veya apoptotik etkilerinin araştırılmasıdır. Bu amaçla, Skov-3 insan over adenokarsinoma hücre soyundaki hücreler, McCoy besiyerinde çoğaltıldı ve 6'lı well platelere ekildi. Arı sütünün stok solüsyonu hazırlandı (1000mg arı sütü 10ml distile suda çözdürülerek) ve hazırlanan stok çözeltiden 1mg/ml, 5mg/ml, 10mg/ml, 20mg/ml ve 50mg/ml arı sütü dozları medyum içine eklenerek 24, 48 ve 72 saat boyunca inkübe edildi. Arı sütü uygulaması sonrasında, yapılan hücre canlılık testini (Tripan Blue) takiben, arı sütünün proliferatif etkisini belirlemek amacıyla Ki-67, apoptotik etkisini belirlemek amacıyla aktif caspase-3 ve aktif PARP-1R ifadeleri immunositokimyal ve immunfloresan yöntemle incelendi. Ayrıca apoptozisin belirlenmesi için kullanılan TUNEL yöntemi ile immunositokimyasal ve immunfloresan bulgular desteklendi. Deneylerin sonucunda, arı sütünün 1mg/ml ve 24 saat uygulamasının hücre proliferasyonu ve apoptozis üzerine etkili olmadığı, ancak genel olarak 50 mg/ml'lik 72 saat arı sütünün kanser hücrelerinde proliferasyonu inhibe ettiği, apoptozisi ise indüklediği belirlendi. Sonuç olarak, ovaryum kanseri üzerine alternatif bir tedavi olarak arı sütünün, gerek monoterapik gerekse kombine kullanımının yeni deneysel protokollere temel oluşturabileceği düşünülmektedir.Item Effects of capsaicin on nitric oxide synthase isoforms in prepubertal rat ovary(Taylor & Francis, 2012-04) Altunbaş, Korhan; Tütüncü, Şerife; Özden, Osman; Sevimli, Alper; Zik, Berrin; Peker, Sabire; Akkoç, Cansel Güzin Özgüden; Uludağ Üniversitesi/Veteriner Fakültesi/Histoloji-Embriyoloji Anabilim Dalı.; F-2822-2017; AAH-9810-2021; 6507763192; 55109615900; 36138936300Nitric oxide (NO) has emerged as an important intra-ovarian regulatory factor. We investigated effects of low dose capsaicin (CAP) treatment on the different NOS isoforms in prepubertal rat ovaries. Fifteen 21-day-old female Sprague-Dawley rats were divided randomly into three groups. The first group received no treatment, the second group received 0.5 mg/kg/day CAP dissolved in the vehicle, and the third group was treated with the vehicle only. The animals were euthanized by ether inhalation after 15 days and their ovaries were excised. Ovaries were fixed in 10% neutral buffered formalin and embedded in paraffin. Sections were processed for standard immunohistochemistry using the labeled streptavidin-biotin technique for expression of nNOS, eNOS and iNOS. We demonstrated that CAP induced expression of NOS isotypes including eNOS, iNOS and nNOS in prepubertal rat ovaries. CAP may lead to release of NO either directly from nerves or indirectly by evoking release from other cells via the action of neuropeptides that are released from afferent terminals and are involved in regulating female reproductive function.Item Histological investigations on the effects of feeding a diet containing red hot pepper on the reproductive organs of the chicken(Wiley, 2005-06) Özer, Aytekin; Erdost, Hatice; Zik, Berrin; Uludağ Üniversitesi/Veteriner Fakültesi.; AAH-9810-2021; AAH-9216-2021In the present study the growth and reproductive organ differences in chickens fed a diet containing 1% red hot pepper (10 g/kg diet) from the first day of age were investigated. In birds fed with the experimental diet it was observed that the abdominal fat content decreased. During the experiment the increase in weight gain in the treated group in the first 4 months was reversed in favour of the control group in month 5. Follicular development in the treated group was faster and laying started 11 days before the control group, and the epithelial and muscular development of the oviduct was always greater than that of the control group. The results indicated that red hot pepper consumed in lower concentrations during the development period in the chickens caused faster development of the reproductive system organs. Laying started 11 days earlier in chicks fed with the red hot pepper added diet, an important economic aspect for egg producers, but which may have implications for other animals. A decrease in abdominal fat content and disorders of lipid metabolism are still under investigation.Item Horozlarda acı kırmızı biberli rasyonla beslemenin üropigi bezi üzerine etkisinin histolojik yönden i̇ncelenmesi(TÜBİTAK, 2002) Zik, Berrin; Erdost, Hatice; Uludağ Üniversitesi/Veteriner Fakültesi/Histoloji ve Embriyoloji Anabilim Dalı.; AAH-9810-2021; AAH-9216-2021; 6507763192; 6505787570In the present study, differences occurring in the uropygial glands of roosters fed with a diet containing red hot pepper from 1-day old until 5-months old, were investigated in their histological and morphometrical aspects. It was determined in the experimental group that the lipid content of cells in the sebaceous zone of the uropygial gland was more than that of the controls, and the thicknesses of the tubulus walls and the length of the tubulus increased depending on both age and the effect of red hot pepper. It was observed that in the cells of the glycogen zone: while glycogen and neutral mucins were abundant in the control group, acid mucins were abundant in the experimental group. At the same time, the thicknesses of the tubulus walls and the length of the tubulus increased depending on age in the control group, but in the experimental group the length of the tubulus increased depending on age, while red hot pepper had a reductive effect on the thicknesses of the tubulus walls. In conclusion, in the experimental group, an increase was observed in the activity of the uropygial gland and this increase was in favour of lipid secretion, on the other hand red hot pepper had an enhancing effect on the secretion of acid mucins and a lowering effect on the secretion of glycogen and neutral mucins.Item Immunohistochemical localization of insulin like growth factor-I receptors in the mice intestine(Indian Veterinary Journal, 2005-06) Yağcı, Artay; Zik, Berrin; Altunbaş, Korhan; Uludağ Üniversitesi/Veteriner Fakültesi/Histoloji-Embriyoloji Anabilim Dalı.; AAH-9810-2021; 6507763192; 8615565500The objective of the present study was to determine the distribution of IGF-I receptors in the adult mice intestinal tract and the differences among trie regions of the intestine. An immunohistocehmical method was adopted to define the cellular distribution of IGF-IR in the intestional tracat. IGF-IR was found at a greater density in the crypt epithelium of the small intestine, The straining was remarkably less both in the apical region of the villi and in the crypt epithelium of the colon. In addition to the canonical pathway involving SMAD proteins, the mitogen-activated kinase (MAPK) pathway via extracellular signal-regulated kinases 1/2 (ERK1/2) is also involved in promoting and maintaining a mesenchymal phenotype by tumor cells following TGF beta signal activation. As dual-specificity phosphatases (DUSPs) regulate ERK1/2 activity by dephosphorylation, we aimed to examine DUSPs' expression upon TGF beta stimulation and whether DUSPs play a role in the EMT and related phenotypes promoted by TGF beta 1 in A549 cells. We found that TGF beta 1 stimulation led to marked changes in several DUSP proteins, including significant decreases in DUSP4 and DUSP13 expressions. We then showed that the ectopic co-expression of DUSP4/13 suppresses TGF beta 1-induced ERK1/2 phosphorylation and protein levels of the EMT transcription factors Snail and Slug proteins. We then demonstrated that DUSP4/13 co-expression partially inhibited TGF beta 1-promoted migration, invasion, and chemoresistance in A549 cells. Insulin-like growth factor-I (IGF-I) is expressed in granulosa cells and has an important role in ovarian development. However, there are no data about the IGF-I expression in ovarian granulosa cells after capsaicin treatment. The aim of this study was to investigate the expression of IGF-I and its receptor (insulin-like growth factor-I receptor [IGF-IR]) in primary rat ovarian granulosa cells after low and high doses of capsaicin treatment. For this, granulosa cells were isolated and cultured from ovaries of 30-day-old female Sprague-Dawley rats. Granulosa cell plates were divided into four groups as cell control (C), vehicle control (V), and 50 mu M and 150 mu M capsaicin groups. In experimental groups, granulosa cells were exposed to capsaicin for 24 hours and immunocytochemistry was performed afterwards using anti-IGF-I and anti-IGF-IR antibodies. Both IGF-I and IGF-IR expressions were found to be significantly increased in parallel to the capsaicin doses. Elevated levels of IGF-I may be a risk factor for ovarian development. Our goal was to demonstrate the occurrence of apoptosis and the cessation of cell proliferation at every phase of scheduled chemotherapy for naturally occurring TVT. Tissue samples were collected immediately after weekly treatments with vincristine sulfate and processed for histological purposes. Sections 5 mu m thick were stained by the TUNEL reaction for apoptosis and immunostained for Ki67 as a proliferation marker. We observed that after vincristine applications, tumor cell proliferation ceased and apoptosis increased. Ki67 HSCORE values were significantly lowered after the first and second treatments with the chemotherapeutic agent compared to controls, whereas TUNEL HSCORE values were significantly higher after two applications of vincristine compared to controls. Our results suggest that scheduled vincristine sulfate applications stabilize the induction of tumor regression by inducing apoptosis and preventing cell proliferation.
