Antioxidant capacity, bioaccessibility of phenolics, hydrolysable and extractable phenolics, sugar, phenolic and fatty acid profiles were determined in the Spirulina honey. Total phenolic content of the Spirulina honey was 14.37 mg GAE/100g and increased 28.53% with Spirulina enrichment. Antioxidant capacities of the Spirulina honey were determined as 6.31, 5.85 and 3.66 mu mol Trolox/g by ABTS, CUPRAC and DPPH methods in bioaccessible extracts. No significant difference was observed between control and Spirulina honey samples in terms of bioaccessibility of the total phenolics (p <= 0.01). Among the detected phenolic compounds (15 phenolics), acacetin (48.55%) and pinocembrine (47.13%) were the highest. Total phenolic content of the honey can be enriched up to 20% by feeding of the honey bees with 10% of Spirulina. Sexually mature drones were used for semen collection. Pooled semen was diluted with TL-Hepes based extender supplemented with different concentrations of BSA (1 mg/mL, 3 mg/mL, and 5 mg/mL) or without BSA (control), at a final concentration of 100x10(6) spermatozoon/mL. Motility, plasma membrane functional integrity (HOST), and defected acrosome (PSA-FITC) were evaluated in the study. At post thaw, the highest sperm motility rates were obtained in the BSA5 group (P< 0.05). Functional integrity of sperm membrane was better preserved in the BSA3 and BSA5 groups compared to the other groups. The acrosomal integrity rates were higher in BSA5 group than in control group (P< 0.05). The study shows that bovine serum albumin supplemented TL-Hepes based extenders have beneficial effect on drone semen parameters at post-thaw. In the present study, we investigated the chronic exposure of chemicals set (deltamethrin, acetamiprid, oxalic acid, fumagillin, and amitraz) on some sperm parameters and on the histomorphology of seminal vesicles. We also assessed the colony development and nosema load before and after the exposure. Thirty native Apis mellifera anatolica honeybee colonies with sister queens equalized with brood and total frame of bees were used for this experiment. Five colonies were used for each group. Deltamethrin, acetamiprid and fumagillin were given as oral chronic exposure at final concentrations of 25.10(-6) mg L-1, 0.01 m L-1 and 50 mg L-1 respectively in syrup solution (50/50). Colonies were exposed to oxalic acid by spraying 5 mL per frame space of 3% (w/v) of oxalic acid dihydrate. Finally, the amitraz was applied based on the manufacturer's instructions. The concentrations chosen represented the field-realistic concentrations and those used by beekeepers in the region. Results showed that deltamethrin reduced brood production. In the same group, we found a high increase in nosema load. All treatments decreased sperm count except for fumagillin but this compound increased sperm mortality and increased the percentage of sperm with defected acrosome integrity. The amitraz exhibited a high sperm mortality and high percentage of sperm with defected membrane integrity function. The sperm parameters such as the count, the motility, the acrosome integrity, the membrane function of sperm, and the histomorphology of seminal vesicles of drones exposed to oxalic acid were the most affected. Semen was obtained from mature drones (16 days or older) harvested from four colonies. Collected semen was diluted to a final concentration of 1/5 (semen/extender) in 0% cryoprotectant (control), 6% glycerol, 6% Ethylene Glycol, 6% 1,2 propanediol or 6% DMSO using a two-step dilution method. The equilibrated semen was frozen in 0.25-ml straws. The percentage of sperm motility and swollen tails (HOST) spermatozoa were evaluated following dilution with extender A (non-cryoprotectant), equilibration and post-thaw stages. In terms of post-thaw motility and plasma membrane integrity recovery we can rank the used cryoprotectant as DMSO, Ethylene Glycol, Glycerol and 1,2 Propanediol; respectively. We determined that 19 apiaries were infected and the suspected clinical signs of foulbrood diseases were investigated in 102 beehives by PCR and cultural method. Broods and combs from colonies with suspected clinical symptoms of foulbrood diseases were collected and cultured for bacteriological examination. All of the specimens contaminated with bacteriae and 37 species of bacteriae were isolated such as Stapylococcus epidermidis, Bacillus subtilis, Corynebacterium jeikum, Corynebacterium pseudotuberculosis, Bacillus spp. All of these bacteria are related to human, animal and environmental origins. In this study, Paenibacillus larvae by PCR amplifying the 973-bp region PL1 and PL2 with 1f, Melissococcus plutonius amplifying the 973-bp region EFB-F and EFB-R gene were amplified. American Foulbrood causative agent Paenibacillus larvae and European Foulbrood causative agent Melissococcus plutonius were not detected in any sample examined by PCR and cultural methods. On the other hand, Paenibacillus alvei that is a seconder agent to European Foulbrood was found in two samples by cultural methods. In conclusion, the results showed that P. larvae and M. plutonius are not present in South Marmara Region. Two groups of honey bee colonies Apis mellifera anatoliaca were used: one as experimental (20) and the other control (10), which were equalized in terms of the number of brood frames, frames with bees, food, hive materials, and health treatments. The experimental colonies were heated during the cold season, and the temperature and humidity inside the hive were recorded. The number of frames of brood, frames of bees, and Varroa destructor mite numbers were also determined for all hives. Results suggest that the number of brood frames and frames with bees increased significantly in the hives with the heating treatment. However, varroa mite numbers also increased significantly in the heated colonies in comparison to the control colonies. Colony losses were significantly less (15%) in heated colonies compared to a 40% loss in control colonies. Semen samples obtained from sexually mature drones were pooled. Then, pooled semen was diluted with different concentrations of L-carnitine (2.5 mM/5 mM/10 mM) and without L-carnitine (control) supplemented extenders. Motility, plasma membrane functional integrity, acrosomal integrity and mitochondrial function were negatively affected by the cryopreservation process but DNA integrity was not affected. Malondialdehyde (MDA) concentration was used for analyzing the membrane lipid peroxidation status and a better result was obtained in the LC5 group compared with the control group at post-thaw. L-carnitine groups had a positive effect on sperm motility, plasma membrane integrity, and mitochondrial function compared to the control group (P<0.05). Acrosomal integrity was better preserved in the LC5 group compared to the control group. The study shows that LC supplemented extenders have beneficial effects on honey bee drone sperm parameters. The results of the present study demonstrated the beneficial effect of using a 5 mMol LC supplemented extender.
