    Evaluation of passive transfer in goat kids with brix refractometer and comparison with other semiquantitative tests

    The aim of this study was to evaluate the passive transfer (PT) status of goat kids by Brix refractometry and compare the results with other semiquantitative tests (total protein - TP, glutaraldehyde coagulation test - GCT, and gamma-glutamyltransferase - GGT) and immunoglobulin G (IgG). The study was conducted on 75 goat kids born from 47 Saanen goats. The blood samples were collected from the kids on day 0 (presuckling) and on the 1st, 2nd, and 3rd days after birth. The Brix% and TP concentrations were measured with refractometers, and GGT activity was measured using a dry chemistry system. The duration of the GCT was determined in the first 60 min. The serum IgG concentration was measured by goat IgG ELISA kit. On the 1st and 2nd days, serum Brix% in the kids was measured as 9.33 +/- 0.17% and 9.17 +/- 0.14%, respectively. In the 1st and 2nd day serum samples of the kids, IgG was 817.76 +/- 37.34 mg/dL and 1173.29 +/- 47.81 mg/dL, respectively, and GGT was 1298.07 +/- 133.29 IU/L and 692.26 +/- 79.86 IU/L, respectively. The Brix refractometer was found to be more sensitive for detection of PT status in kids on the first and second days after birth, such as TP and GCT, whereas GGT, as an early indicator of PT, was useful only on the first day after birth. We conclude that the Brix refractometer could be used to determine the PT status in goat kids and Brix measurements lower than 8.6%, 9.2%, and 9.3% indicate failure of PT in 1-, 2-, and 3-day-old kids, respectively.

    Evaluation of the differences in proteomics of high-quality bovine colostrum and low-quality bovine colostrum

    BackgroundAlthough there are studies on colostrum and milk proteomics of different species in the literature, there is no published report about different quality bovine colostrums' proteomics.ObjectivesThe aim of this study was to compare the proteome content of high- and low-quality bovine colostrums for the first time.MethodsColostrum samples were collected from 32 Holstein cows from the same farm that had just calved. Brix% levels of colostrums were measured, and then, those with a Brix% value of & GE;27% were classified as high-quality and those with a Brix% value of <22% as low-quality. Three samples from high-quality and low-quality colostrums were selected and proteomic analyses were performed by pooling separately.ResultsTotally 95 proteins were identified in the colostrums, and 19 of them showed significant changes between high- and low-quality colostrums. Expressions in colostrum of glycosylation-dependent cell adhesion molecule-1, cofilin-1, alpha-S2-casein, alpha-lactalbumin, alpha-1B-glycoprotein, actin_cytoplasmic-1, nucleobindin-1, cathelicidin-4, inter-alpha-trypsin inhibitor heavy chain H4, chitinase-3-like protein 1 and monocyte differentiation antigen CD14 were lower, whereas tetranectin, secreted frizzled-related protein-1 (SFRP1), perilipin-2, coatomer subunit epsilon (COPE), butyrophilin subfamily 1 member A1, polyubiquitin-B, lactadherin and albumin levels were higher in high-quality colostrum than low-quality colostrum. Moreover, SFRP1, COPE and cathelicidin-4 proteins were identified for the first time in bovine colostrum. In high-quality colostrum, the most prominently down-regulated proteins were cathelicidin-4 (26.01-fold) and cofilin-1 (17.42-fold), and the most prominently up-regulated proteins were COPE (3.37-fold) and tetranectin (3.07-fold).ConclusionsIt was detected that the proteome contents of high- and low-quality bovine colostrums were different from each other. As new functions are added to the protein databases regarding these proteins detected in colostrums, the interactions of proteins with each other and with other molecules will be detailed and the effects of high-quality colostrums on passive transfer immunity and calf health will be understood in full detail.In the presented study, proteomic differences between high- and low-quality colostrums were investigated for the first time. Totally 95 proteins were identified in the colostrums, and 19 showed significant changes between different quality colostrums. Significantly changing proteins have been found to be involved mainly in biological regulation, organization of cellular components, biogenesis, cellular, developmental, immune system, metabolic, multi-organism, multi-cellular processes, localization, locomotion, stimulus-response and signalling pathways. image

    Evaluation of serum choline along with, some biochemical and clinical parameters in cattle suffering from with botulism

    Botulism is a disease of cattle that causes significant impact due to its high mortality rate. The aim of the present study is to evaluate serum choline levels as well as clinical and biochemical parameters of cattle suspected to be suffering from botulism and to compare the results with healthy cattle and also to compare the results of survivors and fatalities. Thirteen botulism suspected and eleven healthy cattle were used. Total protein, albumin, aspartate aminotransferase (AST), alanine aminotransferase (ALT), gamma glutamyl transferase (GGT), calcium, phosphorus, magnesium and potassium levels were measured from the sera of botulism and control cattle by colorimetric method using an autoanalyser. Choline levels were analyzed using HPLC. Dysphagia, decreased tongue strength weak anal reflexes, tongue and tail paralysis and locomotion disorders were the main clinical findings observed in suspected botulism cases. Botulinum toxin analysis of ruminal fluid samples were positive in two cattle from one herd. AST, ALT, potassium and choline levels were higher in cattle suffering from suspected botulism cases and choline levels were positively correlated with albumin and total protein levels. Three animals from botulism group survived. Clinical findings were milder, animals were not recumbent and choline levels were lower in survivors. Clinical findings of the three animals that recovered were milder, animals were not recumbent and choline levels were lower in survivors. It was observed that clinical findings such as decreased tongue strength and anal reflexes, dysphagia, tail paralysis, and locomotion disorders are important for diagnosis of botulism In conclusion, cattle with milder clinical signs and lower choline have a higher chance of survival.

    Effects of vitamin d administration at the beginning of lactation in dairy cows on inflammatory response and liver metabolism

    This study aimed to investigate the effect of vitamin D administered 24 h after calving of dairy cows on calcium-phosphorus metabolism, carbohydrate, lipid and liver metabolism, and inflammatory response. Twenty multiparous Holstein cows were randomly assigned to the experimental (EG) and control group (CG). The EG received 5,000,000 IU vitamin D 3 (5 mL) 24 h after parturition intramuscularly, whereas the CG group received 5 mL of 0.9% NaCl. Blood samples were taken 24 h after parturition before treatments and at the 3rd, 7th, 14th, and 28th days of lactation. Serum NEFA, BHBA, haptoglobin, serum amyloid A (SAA), ceruloplasmin, calcium, phosphorus, parathormone, total protein, triglyceride, AST, and GGT were measured. In addition, total bilirubin, albumin, total cholesterol, and vitamin A concentrations were also measured to calculate liver activity index and the liver functionality index. Subclinical hypocalcemia (SCH) was observed in only one cow in the EG and five in the CG after the 3rd day postpartum. NEFA levels in the EG significantly differed on days 7, 14, and 28 when compared to vitamin D pretreatment values. However, NEFA levels in the CG differed only between day 28 postpartum and the 1st day (p < 0.05). Negative correlation was determined between vitamin D and NEFA (p < 0.01). Conversely, negative correlations were observed between calcium and NEFA, haptoglobin, SAA (p < 0.05). In conclusion, it can be suggested that vitamin D administration can prevent SCH after the 3rd day and has limited positive effects on postpartum NEFA levels.

    Early clinical predictors of diarrhea in the first fifteen days in dairy calves and their relationship with passive transfer status

    The neonatal period is very important in calves. It is extremely important to closely monitor newborn calves with good colostrum management during this period. In this study, the aim was to investigate whether the clinical parameters in the first 15 days after calves are born are correlated with diarrhea and whether those values are the early clinical predictors of the diarrhea that will occur in this period and also to investigate the correlation between diarrhea and the failure of transfer of passive immunity (FTPI). Eighty-two Holstein calves were used in the study. The calves were examined systemically every day and were scored according to a standard form. In the first 15 day period, 64.63% of the calves had diarrhea. In diarrheic calves, the most common findings on the previous day without diarrhea were fetid feces (43.40%), changes in the consistency and color of feces, and a decrease in the temperature of the ears (35.85%). According to non diarrheic calves, fetid feces (P < 0.01), changes in the consistency and color of feces, and nasal discharge were more common among diarrheic calves on the day before diarrhea (p < 0.001). The duration of diarrhea in calves with FTPI was longer than without FTPI. As a result, it is important for calves to be monitored daily in the first 15 days, looking for early signs of diarrhea in this period, namely fetid feces and changes in the consistency and the color of the feces. In addition, coexistence of any fecal changes with the decrease in the temperature of the ears may be clinical predictors of diarrhea. As a result of the daily monitoring of the calves, although the morbidity rate was high, none of the animals died.

    Evaluation of the efficacy of cow colostrum in the treatment and its effect on serum proteomes in calves with cryptosporidiosis

    This study aimed to investigate the effect of using high-quality colostrum in addition to paromomycin on the treatment outcomes and serum proteomes of calves naturally affected by cryptosporidiosis. Thirty Holstein calves infected with only Cryptosporidium spp. were divided into three equal groups. Calves in the PC group received paromomycin orally at a dose of 100 mg/kg once daily for 5 days. Calves in PCOL and PBCOL groups received 250 ml colostrum 3 h after feeding twice a day for 3 days. The PBCOL group was also given 6 g of sodium bicarbonate 15 min before colostrum administration. While the fecal scores of all calves were evaluated daily for 10 days from the initiation of the treatment, fecal oocyst counts were determined on the 0, 1, 2, 3, 5, 7, and 10th days. Brix%, total protein (TP), immunoglobulin G (IgG), gamma-glutamyltransferase (GGT) levels, and proteomic analyses were performed on the 0 and 3rd days' sera. Considering pretreatment values, fecal scores (8th, 2nd, and 2nd day), and fecal oocyst counts (10th, 3rd, and 2nd day) improved in a significantly (p < 0.05) shorter time in the colostrum groups than in the control group. By serum proteomic analysis, 99, 93, and 83 proteomes were detected in PC, PCOL, and PBCOL groups, respectively. Although the significant changes in any protein in Group PC were absent, significant changes were observed in Alpha-1B-glycoprotein (A1BG), Zinc transporterZIP11 (S39AB), Cathelicidin-1 (CTHL1), Actin_ cytoplasmic-1 (ACTB), and Apolipoprotein A-IV (APOA4) proteins in Group PCOL and Alpha-1-antiproteinase (A1AT), Serum amyloid A protein (SAA), Actin-cytoplasmic-2 (ACTG), Protein HP-20 homolog (HP20) proteins in Group PBCOL with colostral treatment, which indicated that the use of colostrum had an effect on calf serum proteomes. The more pronounced healing and shorter clinical improvement time in the colostrum groups especially colostrum with sodium bicarbonate revealed that these proteomes have positive effects in the treatment with their systemic and local effects in the intestines.

    Comparing the effect of ammonium molybdate versus ammonium molybdate and menbutone on hepatic functions of sheep with subclinical copper poisoning

    This study aimed to investigate the effect of using menbutone in addition to ammonium molybdate on liver enzymes in sheep naturally poisoned with copper. Merino lambs (n = 30), naturally poisoned with copper and which also had high liver enzyme levels, were divided into two groups, each with 15 lambs. The AM + MEN group received ammonium molybdate and menbutone and the AM group received only ammonium molybdate solution. Both groups received 1.7% ammonium molybdate solution (1 mL per 10 kg body weight [BW]) subcutaneously on 0, 2nd and 4th days of the study. Menbutone (Genabil (R), Boehringer Ingelheim, Germany) was administered intramuscularly at a dose of 10 mg/kg BW on days 0 and 2, in addition to ammonium molybdate in the AM + MEN group. Blood samples were collected on days 0 and 7, and aspartate aminotransferase (AST), gamma-glutamyltranspeptidase (GGT) and creatinine levels were evaluated. Over 7 days, AST levels decreased from 351.04 +/- 63.50 IU/L to 286.40 +/- 55.68 IU/L in the AM group (P > 0.05) and from 425.00 +/- 119.25 IU/L to 240.83 +/- 29.62 IU/L in the AM + MEN group (P <= 0.05). GGT levels decreased from 121.16 +/- 15.88 IU/L to 110.39 +/- 10.13 IU/L in the AM group (P > 0.05) and 124.52 +/- 15.50 to 98.60 +/- 9.08 IU/L in the AM + MEN group (P <= 0.05). Based on these findings, the use of menbutone, in addition to ammonium molybdate, has significantly reduced the level of liver enzymes.

    Butaphosphan and cyanocobalamin treatment of pregnant ewes: Metabolic effects and potential prophylactic effect for pregnancy toxaemia

    The effect of administration of butaphosphan and cyanocobalamin on the prophylaxis of pregnancy toxaemia in ewe was investigated. Moreover, the effects of these compounds on body weight gain and biochemical metabolism of pregnant ewe was assessed. A total of 59 pregnant Kivircik crossbred ewes were used in this study. Group I (n: 15) was administered butaphosphan and cyanocobalamin three times before delivery at 1-week intervals. Group II (n: 15) was administered butaphosphan and cyanocobalamin three times before delivery at 3-day intervals. Group III (n: 15) was administered 0.9% NaCl three times before delivery at 1-week intervals. Group IV (n: 14) was administered 0.9%NaCI three times before delivery at 3-day intervals. Six blood samples Were taken from each ewe four times before delivery and two times after delivery. Haematological and biochemical analyses were performed.The levels of BHB and NEFA in groups administered butaphosphan and cyanocobalamin were noticeably lower but there were no statistically significance. Elevated BHB (>0.8 mmol/L), subclinical pregnancies toxaemia were identified in 56.66% in test groups and 72.41% in control groups in all ewes and this was higher in the ewes bearing multiple pregnancies 71.42 in test groups and 82.35% in control groups. Subclinical pregnancy toxaemia in pregnant ewes with twins or triplets is lower than the levels for the control groups, despite the greater lamb counts and weights of the ewes in test groups.Based on our results, it was concluded that the butaphosphan and cyanocobalamin combination could be used as an alternative treatment for the prevention of pregnancy toxaemia.
