(Hypericaceae, Clusiaceae) is an endemic species to Turkey, which is one of the commonly used species in pharmacological research. We studied seed germination characteristics. The effects of dry storage (3, 9, 12 month), moist chilling (3, 6, 9, 12 month), scarification (H2SO4, sand paper), tap water, hormone (250, 500, 1000 ppm GA(3)), different temperature (15/10, 20/10 and 25/15 degrees C) and light (photoperiod, darkness) treatments on seed germination of H. adenotrichum seeds collected from subalpine belt of Uludag Mountain were examined. Favourable germination temperature for H. adenotrichum is 20/10 degrees C and no light is required for the seed germination. Seeds of species have physiological dormancy. GA(3) without any other treatment was not successful enough to break the dormancy status alone. Scarification by sulphuric acid and 1000 ppm GA(3) combination is the most effective way for germination of H. adenotrichum seeds. In this study, we investigated the effects of GA(3), the combination of hormone series and short-term moist chilling (1-month), and long-term moist chilling (4-month) on the germination percentage and mean germination time in relation to seed dormancy breaking. All treatment series were incubated under continuous dark (20 degrees C, 24 h) and light/dark (20/10 degrees C, 12/12 h) conditions. Seeds were collected from specimens widespread on the alpine and subalpine grasslands and dwarf shrubs of Mount Uludag (1800-1900 m a.s.l.). Depending on the concentration, GA(3) and the combination of hormone and moist chilling treatments promoted seed germination in both dark and light/dark conditions. After 4-month-moist chilling treatment, seeds germinated 27% in a light/dark and 80% in a dark regime. Hormone and moist chilling treatments reduced the mean germination time. (Scrophulariaceae) were studied as a candidate plant for remediating the Ni polluted soils. The metabolic responses, such as nitrate assimilation (nitrate reductase and glutamine synthetase activity) and antioxidant system activity [superoxide dismutase (SOD), catalase (CAT) and ascorbate peroxidase (APX) activity], of this species exposed to nickel in Hoagland's nutrient medium were investigated as remediation performance parameters. The accumulation of nickel and the variations in the content of some elements (B, Cu, Fe, Mg, Mn, Mo and Zn) and some growth parameters, such as the water content, biomass production, and contents of chlorophyll and soluble protein, were also examined. The accumulation of Ni in both the roots and leaves varied depending on the exposure times and doses. Increased oxidative stress was suggested by the increases in the activities of SOD, CAT and APX. Although some element contents were inhibited by Ni treatments, these inhibitory effects was decreased depending on the time, and even these elements are accumulated in roots. It has the potential to be an ornamental plant, so it is important to know its germination characteristics and seed morphological characteristics. We evaluated the effects of moist chilling (3 to 12 months) and application of gibberellic acid (GA(3)) (250 to 1000 mg/L-1) on seed germination under two different light regimes (light phase and dark phase) and four temperature regimes (20, 20/10, 25/15, and 15/10 C-degrees). Seeds were collected from the subalpine belt of Mount Uludag at an altitude between 2200 and 2300 m. M. bourgaei seeds were dormant and reached the highest germination percentage after 12 months of moist chilling. GA(3) applications only have a limited effect on the breaking of dormancy in most cases. Maximum germination of 28 percent was obtained by the GA(3) application of 1000 mg/L under dark phase with incubation at 15/10 C-degrees. Scarification with sulfuric acid did not result in any germination. Seed germination of above 80 percent was obtained after 6 months of moist chilling and above 90 percent after 9 and 12 months of moist chilling. Intermediate physiological dormancy was determined as the type of dormancy. Our findings on seed germination and dormancy characteristics of M. bourgaei, which is an endemic alpine meadow plant, will contribute to the protection and development of the germplasm of this species.
