Abstract
Organizmada, oksijen reaksiyonlarında elektronu eşlenmemiş atom ve moleküller oluşmaktadır. Serbest radikal olarak adlandırılan yapılar dış orbitallerinde eksik elektron bulundururlar. Serbest radikaller kararsız yapıda bulunurlar. Kararlı yapıya geçmek için moleküllerle reaksiyona girerler. Serbest radikaller aynı zamanda oksidan olarak da adlandırılırlar. Oksidanlar, nükleik asit, lipid ve protein yapılarını hasara uğratarak yapılarına zarar verebilmektedirler. Temel bileşenlerde oluşan hasar ise kanser, bağışıklık sorunları ve tansiyon hastalıklarına sebebiyet verebilir. (Ngo ve diğerleri, 2010). Organizmada oksidanların etkisinden korunmak için gerekli olan antioksidan molekülleri bulunur. Kısaca antioksidan terimini, vücutta üretilen veya gıdalardan sağlanan, hücre hasarını önleyen bileşikler olarak tanımlayabiliriz. Antioksidanlar, doğal olarak vücutta üretilebilirler ya da dışarıdan ek olarak alınabilirler. Algler, günümüzde doğal antioksidan kaynağı olarak sentetiklere kıyasla daha fazla tercih edilebilmektedirler (Gey, 1990). Dasya rigidula (Kützing) Ardissone 1878, (Rhodophyta) Mudanya, Bursa denizin kıyı kesimlerinden kısımlarından toplanmıştır. Dasya rigidula türünün farklı çözücülerle (metanol, %25 metanol, kloroform ve dietil eter) antioksidan potansiyeli belirlenmiştir. Soxhlet kullanılarak ekstraksiyonları yapılmıştır. Evaporatör yardımı ile çözücüler uçurularak özüt eldesi yapılmıştır. Bu özütler kullanılarak türün 2,2-diphenyl-1- picrylhydrazyl (DPPH) radikal süpürücü aktivite, indirgeme gücü, toplam fenol ve toplam flavonoid içerikleri belirlenmiştir. Kloroform ile hazırlanan özütün en yüksek DPPH süpürücü aktivitesine sahip olduğunu görülmüştür. Kloroformla hazırlanmış özüt en yüksek indirgeme gücüne sahiptir. Kloroform ve dietil eterle hazırlanan özütler en yüksek fenolik içeriğe sahiptirler. Kloroform ve dietil eterle hazırlanmış özütlerde en yüksek flavonoid değerleri görülmüştür. Kloroform ve dietil eterle hazırlanmış özütler, metanol ve %25 metanol ile hazırlanmış özütlere oranla yüksek antioksidan etki gösterdikleri gözlemlenmiştir.
In the organism, atoms and molecules with unpaired electrons are formed in oxygen reactions. Structures called free radicals have missing electrons in their outer orbitals. Free radicals exist in an unstable structure. They react with molecules to reach a stable structure. Free radicals are also called oxidants. Oxidants can damage nucleic acid, lipid and protein structures by damaging their structures. Damage to basic components can cause cancer, immune problems and blood pressure diseases. (Ngo et al., 2010). There are antioxidant molecules necessary to protect the organism from the effects of oxidants. Briefly, we can define the term antioxidant as compounds that prevent cell damage, produced in the body or obtained from foods. Antioxidants can be produced naturally in the body or taken externally as a supplement. Algae are now more preferred as a source of natural antioxidants than synthetic ones (Gey, 1990). Dasya rigidula (Kützing) Ardissone 1878, (Rhodophyta) was collected from the coastal parts of the sea in Mudanya, Bursa. The antioxidant potential of Dasya rigidula species was determined with different solvents (methanol, 25% methanol, chloroform and diethyl ether). Extractions were made using Soxhlet. The extract was obtained by evaporating the solvents with the help of an evaporator. Using these extracts, the 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity, reducing power, total phenol and total flavonoid contents of the species were determined. It was observed that the extract prepared with chloroform had the highest DPPH scavenging activity. The extract prepared with chloroform has the highest reducing power. Extracts prepared with chloroform and diethyl ether have the highest phenolic content. The highest flavonoid values were observed in extracts prepared with chloroform and diethyl ether. It was observed that extracts prepared with chloroform and diethyl ether showed higher antioxidant effects than extracts prepared with methanol and 25% methanol.
