Abstract
Bu çalışmada, etlik civcivlerin içme sularına 2 ppm dozunda iyot katılmasıyla tiroid bezlerinde meydana gelen yapısal değişiklikler histolojik yöntemler ile saptanmıştır. Deney grubunun içme suyuna 2 ppm dozunda iyot ilavesi yapılırken, kontrol grubuna iyot katılmamış, her grubun yemlerine de mineral premiksi yoluyla 1 ppm iyot ilave edilmiştir. Etlik civcivlerden tiroid örnekleri 21. ve 43. günlerde alınmıştır. Alınan örnekler histoloji tekniğine uygun işlemlerden geçirilerek parafinde bloklanmıştır. Bloklardan hazırlanan histolojik kesitlerin bir kısmına yapısal özelliklerin belirlenmesi için Crossmon'ın üçlü boyama tekniği, diğer bir kısmına ise kolloid içerisindeki karbonhidratların belirlenebilmesi için Mc. Manus'un Periodic Acide Schiff (PAS) metodu uygulanmıştır. Histolojik ve morfometrik değerlendirmeler ile 21 ve 43 günlük kontrol grubuna verilen iyot miktarının tiroid foliküllerinin normal aktivitesi için yetersiz olduğu saptanmıştır. Deney grubunda ise özellikle 43 günlüklerde, yassı epitele sahip folikül sayılarında azalma buna karşılık kübik ve prizmatik epitel hücreli foliküllerde artış görülmüştür.
This experiment was conducted to investigate structural effects on the thyroid glands of 2 ppm iodine supplementation to the drinking water of the broiler chicken. 2 ppm supplementary iodine was added to the drinking water of the experimental group while the drinking water of the control had no iodine. The feeds of both groups contained 1 ppm iodine originate from premix. Thyroid samples were taken from the broiler chickens on the 21st and 43rd days. Rutine histological methods were applied to the samples and then they were embedded paraffine blocks. Some of the tissue sections were stained by Crossmon's Triple Stain for demonstration of structural praperties and the others by Mc. Manus's Periodic-Acide Schiff (PAS) for demonstration of carbonhydrates in colloid. In the histological and morfometrical conclusions, it was indicated that iodine dose given to the control group at the 21-day-o/d and 43-day-old was not enough for the normal activity of thyroid follicles. Especially in the 43-day-old chicks of the experimental group the number of follicules with simple squamous follicular cells decreased as simple columnar and cuboidal cells increased.
