Abstract
Bu çalışmada, Servikal Mukus Penetrasyon Testi uygulanan 50 adet döl tutmayan inek ve düvenin, servikal mukus ile kan serumlarında aglütinasyon reaksiyonunun saptanması ve anılan olgunun spermatozoon motilitesi ve migrasyon değerleri üzerine olan etkisinin incelenmesi amaçlandı. Deneme grubunu tohumlamada kullanılan, kontrol grubunu ise tohumlamada kullanılmayan dondurulmuş boğa spermaları oluşturdu. Deneme grubunda aglütinasyon reaksiyonu servikal mukus örneklerinin 9’unda (%18) pozitif, 41’inde (%82) negatif olarak saptandı. Kontrol grubunda ise aglütinasyon reaksiyonu servikal mukus örneklerinin hepsinde negatif olarak tespit edildi. Deneme ve kontrol gruplarına ait ortalama motilite ve migrasyon değerleri 9 adet servikal mukus örneğinde %7.22±1.69, %8.89±2.61 ve 1.85±0.22 cm, 3.61±0.29 cm (P<0.01); 41 adet servikal mukus örneğinde ise %8.04±1.16, %11.34±1.56 (P<0.05) ve 3.31±0.23 cm, 3.72±0.22 cm olarak ölçüldü. Ayrıca deneme gruplarına ait migrasyon değerleri istatistiksel olarak önemli bulundu (P<0.001). Deneme ve kontrol gruplarına ait aglütinasyon derecesi, ortalama motilite ve migrasyon değerleri 9 adet kan serum örneğinin 1’inde kuvvetli (%11.11), 2’sinde orta (%22.22), 6’sında normal (%66.66), %31.11±2.86, 0.51±0.07 cm ve 1’inde kuvvetli (%11.11), 2’sinde orta (%22.22), 6’sında normal (%66.66), %27.77±4.86, 0.46±0.08 cm; 41 adet kan serum örneğinin ise 7’sinde kuvvetli (%17.07), 14’ünde orta (%34.14), 20’sinde normal (%48.78), %22.07±1.75, 0.63±0.03 cm ve 8’inde kuvvetli (%19.51), 18’inde orta (%43.90), 15’inde normal (%36.58), %20. 85±1.74 ve 0.69±0.04 cm olarak ölçüldü. Ayrıca deneme gruplarına ait motilite değerleri istatistiksel olarak önemli bulundu (P<0.05). Sonuç olarak, servikal mukusta oluşan aglütinasyon reaksiyonunun spermatozoonların motilitesini ve migrasyon değerlerini olumsuz yönde etkilediğini düşünmek olasıdır. Ayrıca kan serumuna ait bulguların servikal mukus bulgularını destekler nitelikte olmadığı kanısına varılmıştır.
In this study, the purpose was to determine agglutination reaction in cervical mucus and serum samples of 50 repeat breeder cow and heifers on which Cervical Mucus Penetration Test was applied, to investigate the effect of mentioned situation on sperm motility and migration values. Frozen bull sperm used for artificial insemination and never used for artificial insemination formed test and control groups, respectively. In test group, agglutination reaction was positive in 9 samples (18%) and negative in 41 samples (82%) in cervical mucus samples. However, in control group, agglutination reaction was negative in all samples. Average motility and migration values belonging to test and control groups were measured as 7.22%±1.69, 8.89±2.61 and 1.85±0.22 cm, 3.61±0.29 cm (P<0.01) in 9 cervical mucus samples; and 8.04±1.16, 11.34±1.56 (P<0.05) and 3.31±0.23 cm, 3.72±0.22 cm in 41 cervical mucus samples. In addition, migration values of test grups were found statistically important (P<0.001). Agglutination grade, average motility and migration values in 9 blood serum samples were determined as strong in one of samples (11.11%), medium in two of samples (22.22%), normal in six of samples (66.66%), 31.11%±2.86 and 0.51±0.07 cm in test group; and strong in one of samples (11.11%), medium in two of samples (22.22%), normal in six of samples (66.66%), 27.77%±4.86 and 0.46±0.08 cm in control group; in 41 blood serum samples, the same values were found as strong in 7 of samples (17.07%), medium in 14 of samples (34.14%), normal in 20 of samples (48.78%), 22.07%±1.75 and 0.63±0.03 cm in test group; and strong in 8 of samples (19.51%), medium in 18 of samples (43.90%), normal in 15 of samples (36.58%), 20.85%±1.74 and 0.69±0.04 cm in control group.
