Abstract
Normal sağlıklı kişilerden (n=50) elde edilen serumlarda farklı tekniklerle yüksek dansiteli lipoprotein kolesterol (HDL-K) tayini yapıldı: 1- Dekstran sülfat-magnezyum (DS-Mg) presipitasyon, enzimatik yöntemle kolesterol ölçümü (EDS), 2-DS-Mg presipitasyon, kimyasal yöntemle (Zak) kolesterol ölçümü (ZDS), 3- Fosfotungstat-Mg (FT-Mg) presipitasyonu, enzimatik yöntemle kolesterol ölçümü (EFT), 4-FT-Mg presipitasyonu, kimyasal yöntemle kolesterol ölçümü (ZFT). Dört değişik şekilde ölçülen HDL-K düzeylerinin birbirlerinden anlamlı şekilde farklı oldukları ancak, aralarında anlamlı düzeyde korelasyon gösterdikleri saptandı. En yüksek korelasyon (r=0. 705) EFT ile ZDS yöntemleri arasında bulundu. Çalışma-içi değişkenlik katsayısı(% CV) EDS, ZDS, EFT ve ZFT için normal havuzda sırasıyla 9. 71, 5.23, 7.38, 3.69; yüksek havuzda 12.15, 15.36, 10.12 ve 6.20 idi. Günler-arası% CV +4 C'de bekletilen serumlarda, - 20 C'de bekletilenlere göre biraz daha düşük bulundu. Sonuçta, HDL-K tayininde isabet derecesinin iyi olmamasından presipitasyon işleminin sorumlu olduğu düşünüldü.
High density lipoprotein-cholesterol (HDL-C) was determined in the sera of normal healthy persons (n= 50) with different techniques: 1-Precipitation with dextran sulphate-magnesium (DS-Mg), enzymatically choleste of determination (EDS), 2-Precipitation with DS-Mg, chemically (Zak) cholesterol determination (ZDS) 3- Precipitation with phosphotungstate-Mg, enzymatically cholesterol determination (EFT), 4-Precipitation with FT-Mg, chemically cholesterol determination (ZFT). The HDL-C levels found by 4 different methods were significantly different from each other; on the other hand there were significantly correlations among each other. The highest correlation (r=O. 705) was found between EFT and ZDS nı thods. Within-run coefficient of variation (%CV) in the normal serum pool for EDS, ZDS, EFT and ZFT were 9. 71, 5.23, 7.38, 3.96; and 12.1 5, 1 5.36, 10.12, 6.2 in the high serum pool, respectively. Between-day% CV was found slightly tower in the sera kept at +4C than the sera kept at - 20C. Finally, we thought that the reason for precision not being satisfactory, is the precipiıation step.
