Bromokriptinin adrenal gland ve simpatetik ganglionlarda tirozin hidroksilaz aktivitesine etkisi

Abstract
Bu çalışmada ergot bileşiklerinden güçlü bir dopaminerjik agonist bromokriptinin adrenal gland ve çok sayıda simpatetik ganglionda tirozin hidroksilaz aktivitesine etkisi incelenmiştir . Dişi sıçanlara 3 gün süreyle mide sondası yoluyla bromokriptin (50 mg/ kg) verilmiştir . Dördüncü gün öldürülen sıçanlardan adrenal gland ve simpatetik ganglionlar (2 servikal, 10 torakal, 6 lumbar ve söliak ganglion) çıkarılmış ve bunlarda tirozin hidroksilaz aktivitesi ölçülmüştür. Bromokriptin adrenal glandda tirozin hidroksilaz aktivitesini belirgin olarak (kontrolün % 153) ve anlamlı (p < 0.02) olarak arttırmıştır. Bromokriptin, lumbal bölgedeki ilk üç ganglionda da tirozin hidroksilazı arttırmıştır. Bunlardan yalnız a 2. lumbar ganglion etki anlamı (p < 0.01) ve adrenal glandda gözlediğimiz etki kadardı . Bromokriptin söliak ganglionda etkisiz bulundu. Bromokriptin verilen sıçanlarda tirozin hidroksilaz aktivitesi üst servikal ganglionda (% 121), alt servikal ganglion (% 124) ve 1. torasik ganglionda (% 131) artmış bulundu. Bunun dışında kalan ganglionlarda ise enzim aktivitesi değişmemiş bulundu. Bu bulgular bromokriptinin simpato-adrenal sistemde seçici olarak merkezi yolla uyarılma yaptığını göstermektedir.
In the present study, effects of bromocriptine, a potent dopaminergic agonist, on tyrosine hydroxylase activity in adrenal gland and in various sympathetic ganglia were investigated. Female rats received 50 mg/ kg of bromocriptine, daily for three consecutive days, by stomach tube. Animals were killed 24 hours after the last treatment and their adrenal gland and sympathetic ganglia (2 cervical. 10 thoracal, 6 lumbar and coeliac ganglion) were dissected for measurement of tyrosine hydroxylase activity significantly (p < 0.02), by about 150 % in the adrenal gland. Bromocriptine also increased the enzyme activity in the first three lumbar sympathetic ganglia. The increase was significant (p < 0.01), by about similar extend to the increase observed in the adrenal, in the second lumbar ganglion . The enzyme activity showed a tendency to increase (by about 121 -131 % of the control) in superior and inferior cervical and first thoracic ganglia, but failed to reach significant value. Tyrosine hydroxylase activity was found unaltered in all other sympathetic ganglia in bromocriptine treated animals.These data show that bromocriptine can cause a selective increase in impulse low to the sympatho- adrenal system from central nervous system.
