Sıçanlarda sistemik inflamasyon modelinde A vitamini ve balık yağından zengin beslenmenin ovaryum dokusu üzerine etkisi

Abstract
Ovaryumlarda farklı yolaklar üzerinden gelişen inflamatuar olgularda nutrisyonel desteklerin araştırıldığı çalışmalar bulunmakla birlikte literatürde A vitamini ve balıkyağı kombinasyonuna rastlanmamıştır. Bu çalışmada lipopolisakkarit (LPS) ile indüklenen inflamatuar hasar sonrasında A vitamini ve omega-3 kaynağı olan balıkyağından zenginleştirilmiş beslenme şartlarının sıçan ovaryum dokusu üzerindeki etkilerinin araştırılması amaçlandı. Çalışmada kullanılan Wistar albino dişi sıçanlar: (1) A Vitamini + LPS Grubu, (2) Omega 3 + LPS Grubu, (3) Kombine Yem + LPS Grubu, (4) LPS Grubu ve (5) Kontrol Grubu olarak beş gruba ayrıldı. Sıçanlara 7 gün 0,5 mg/kg/gün LPS intraperitoneal olarak uygulandı. Sonrasında 28 gün süre ile zenginleştirilmiş içerikli beslenmeler uygulandı. Deneklerden alınan ovaryum dokularının hematoksilen-eozin ile boyanmış parafin kesitlerinde histopatolojik ve morfometrik değerlendirmeler gerçekleştirildi. Kan örneklerinde ELISA ile serum TNF-α düzeyleri çalışıldı. Uygulanan LPS’nin sıçan ovaryumunda kronik inflamasyona neden olduğu histolojik ve biyokimyasal olarak gösterildi. Ayrıca folikül gelişimini olumsuz yönde etkilediği, atrezik ve kistik folikül oluşumuna neden olduğu görüldü. A vitamini veomega-3’ten zengin kombine diyetin tekli kullanımlarına göre inflamasyonu baskılamada, atrezik ve kistik folikül gelişimini azaltmada daha etkili olduğu saptandı. Sonuç olarak; nutrisyonel A vitamini ile balık yağının kombinasyon formunda kullanımının ovaryumların inflamatuar olgularında destek tedavi seçeneği olarak kullanılabileceği düşünüldü.
Although there are studies investigating nutritional supplements in inflammatory cases that develop through different pathways in the ovaries, a combination of vitamin A and fish oil has not been found in the literature. This study aimed to investigate theeffects of nutritional conditions enriched with vitamin A and fish oil, a source ofomega-3, on rat ovarian tissue after lipopolysaccharide (LPS)-induced inflammatorydamage.Wistar albino female rats used in the study were divided into five groups: (1)Vitamin A + LPS Group, (2) Omega 3 + LPS Group, (3) Combined Feed + LPS Group, (4) LPS Group and (5) Control Group. 0.5 mg/kg/day LPS was administeredintraperitoneally to rats for 7 days. Afterwards, enriched diets were administered for28 days. Histopathological and morphometric evaluations were performed on paraffin sections stained with hematoxylin-eosin of ovarian tissues removed from the rats. Serum TNF-α levels were measured in blood samples by ELISA.It has been shown histologically and biochemically that LPS causes chronicinflammation in the rat ovary. It was also observed that it negatively affected follicledevelopment and caused the formation of atretic and cystic follicles. It was determinedthat the combined diet rich in vitamin A and omega-3 was more effective insuppressing inflammation and reducing the development of atresia and cystic folliclesthan their single use.In conclusion; it was thought that the use of nutritional vitamin A and fish oil in combination form could be used as a supportive treatment option in inflammatory cases of the ovaries.
