SD-OCT analysis of the macula and peripapillary RNFL was obtained for each participant. In addition, patient demographics and comorbidities were recorded.Results: 238 eyes of 122 subjects (Group 1: n = 63; Group 2: n = 59) were included. The incidence of coexisting comorbidity was higher in Group 1 (n = 26/63, 41.3%) compared with Group 2 (n = 12/59, 20.3%) (p = 0.013). The central foveal thickness (CFT) was significantly higher in Group 1 (271.0 +/- 26.8 mu m) than Group 2 (263.2 +/- 22.0 mu m) (p = 0.015). The average outer nuclear layer (ONL) thickness at central fovea in Group 1 (85.4 +/- 13.3 mm) was significantly thicker than that in Group 2 (81.4 +/- 15.2 mu m) (p = 0.035). The mean peripapillary RNFL thickness of Group 1 (102.6 +/- 8.8 mu m) and Group 2 (100.9 +/- 8.3 mu m) were similar (p = 0.145). The mean choroidal thickness of groups at the fovea and at 1500 mu m nasal and temporal to the fovea were not significantly different (p> 0.05 for all).Conclusion: Significant thickness alterations in individual retinal layers and CFT was detected in post-COVID-19 patients. However, the number of patients on cadaveric kidney waiting lists is increasing. This situation also makes it difficult to follow cadaveric kidney recipient candidates at transplantation centers. It is very important for the medical status and information of the waitlisted candidates to be current at the time of the cadaveric kidney transplant call. In this study, we investigated the exclusion reasons of cadaveric transplantation candidates at our center.MATERIAL and METHODS: A total 252 adults called for cadaveric kidney transplantation between January 2013 and August 2015 were included the study. Electronic data, immunology laboratory results and transplantation council records were examined. Exclusion reasons of the candidates were evaluated according to the list order. Candidates with a lower rank than the transplanted candidate were excluded.RESULTS: In this period, 252 (56.7% male, 43.3% female, 46.2 +/- 12.2 years) adults were called for 81 cadaveric donors (120 kidneys). A total of 144 (57.1%) candidates were excluded. Exclusion reasons were being unable/unwilling to come to the center (59, 34.1%), positive lymphocyte cross-match test (40, 22.9%), inability to reach the patient (24, 13.9%), chronic HCV infection (20, 11.6%), active infection (11, 6.6%), cardiovascular reasons (8, 4.6%), and respiratory system disorders (5, 2.5%).CONCLUSION: Unexpected problems of cadaveric donor candidates can lead to exclusion from transplantation. The study population comprised 322 children diagnosed with multisystem inflammatory syndrome.ResultsThe organ systems most commonly involved were the cardiovascular and hematological systems. Intravenous immunoglobulin was used in 294 (91.3%) patients and corticosteroids in 266 (82.6%). Seventy-five (23.3%) children received therapeutic plasma exchange treatment. Patients with a longer duration of the PICU stay had more frequent respiratory, hematological, or renal involvement, and also had higher D-dimer, CK-MB, and procalcitonin levels. A total of 16 patients died, with mortality higher in patients with renal, respiratory, or neurological involvement, with severe cardiac impairment or shock. The non-surviving group also had higher leukocyte counts, lactate and ferritin levels, and a need for mechanical ventilation.ConclusionsIn cases of MIS-C, high levels of D-dimer and CK-MB are associated with a longer duration of PICU stay. Non-survival correlates with elevated leukocyte counts and lactate and ferritin levels. We assessed real-life 6- and 12-month effectiveness (i.e., retention, remission, low disease activity [LDA], and response rates) of the IL-17 inhibitor secukinumab in PsA patients overall and across 1) number of prior biologic/targeted synthetic disease-modifying antirheumatic drugs (b/tsDMARDs), 2) years since diagnosis, and 3) European registries. Methods Thirteen quality registries in rheumatology participating in the European Spondyloarthritis Research Collaboration Network provided longitudinal, observational data collected as part of routine care for secondary use. Data were pooled and analyzed with Kaplan-Meier plots, log rank tests, Cox regression, and multiple linear and logistic regression analyses. Results A total of 2,017 PsA patients started treatment with secukinumab between 2015 and 2018. Overall secukinumab retention rates were 86% and 76% after 6 and 12 months, respectively. Crude (LUNDEX adjusted) 6-month remission/LDA (LDA including remission) rates for the 28-joint Disease Activity Index for Psoriatic Arthritis, the Disease Activity Score in 28 joints using the C-reactive protein level, and the Simplified Disease Activity Index (SDAI) were 13%/46% (11%/39%), 36%/55% (30%/46%), and 13%/56% (11%/47%), and 12-month rates were 11%/46% (7%/31%), 39%/56% (26%/38%), and 16%/62% (10%/41%), respectively. Clinical Disease Activity Index remission/LDA rates were similar to the SDAI rates. Six-month American College of Rheumatology 20%/50%/70% improvement criteria responses were 34%/19%/11% (29%/16%/9%); 12-month rates were 37%/21%/11% (24%/14%/7%). Secukinumab effectiveness was significantly better for b/tsDMARD-naive patients, similar across time since diagnosis (4 years), and varied significantly across the European registries. Conclusion In this large real-world study on secukinumab treatment in PsA, 6- and 12-month effectiveness was comparable to that in previous observational studies of tumor necrosis factor inhibitors. Internalized stigma, acceptance of negative attitudes and stereotypes of society regarding a person's illness, has not been studied previously in HS. Objectives: The objective of this study was to investigate the internalized stigma state of HS patients and identify the factors affecting it. Methods: This multicenter, prospective, cross-sectional study included 731 patients. Internalized Stigma Scale (ISS), Hurley staging, Physician Global Assessment, Dermatology Life Quality Index (DLQI), Skindex-16, Beck Depression Inventory-II (BDI-II), and Visual Analog Scale (VAS)-pain score were used in the study. Results: The mean ISS value (57.50 +/- 16.90) was comparable to the mean ISS values of studies in visible dermatological and various psychiatric diseases. A significant correlation was found between the mean values of ISS and all disease activity scores, quality of life measures, BDI-II, and VAS-pain scores. Obesity, family history, low education and income level, vulva/scrotum involvement and being actively treated are significant and independent predictive factors for high internalized stigma in multivariate analysis. Conclusions: HS patients internalize society's negative judgements, which may create a profound negative effect on access to health care. Recently, due to its histological appearance and different involvement areas, the predominated idea is that it is a granulomatous reaction against to the hair follicle destruction and ruptured epidermoid cysts. A 32-year-old female patient admitted to our outpatient clinic because of the acne lesions on her face and hands for 2 months. The patient was diagnosed as LMDF with clinical and histopathologic findings. In addition, biopsy of the papules around the eyelids revealed epidermoid cyst structures associated with the granulomas. On the occasion of this case, we revised LMDF, a dermatosis which still causes different ideas about naming and etiopathogenesis, in the light of the literature; we discussed differential diagnosis and we analyzed the association with epidermoid cysts.Publication Effects of non-genetic factors on growth traits and survival rate in Karacabey Merino lambs(Polska Akad Nauk, 2023-01-01) Oral, H. The study included data from a total of 17659 lambs and 12263 ewes raised in 30 herds between the years 2011 and 2016 as part of the National Sheep and Goat Improvement Project. The aver-age birth weight of the lambs was determined as 3.73 +/- 0.05 kg, the average 45th day live weight was 18.43 +/- 0.58 kg, the average weaning (average 91.8 days) weight was 31.83 +/- 0.24 kg, and the average daily live weight gain until weaning was 289.1 +/- 3.91 g. The average survival rate of lambs at weaning was calculated to be 95.67% +/- 1.15. The effects of the factors herd, birth year, birth type, birth season and sex were found significant for all traits (p<0.01). It was established that the mortality rate in lambs in large herds was higher during 6 years in which the project was carried out. Due to the high twinning rate in large herds, the number of lambs per worker is increased, and as a result, they cannot be adequately cared for. For this reason, large farms may be encouraged to increase workmanship services in addition to being provided with protective health practices for lambs throughout the birth period. On the other hand, it was determined that the twinning rate was low in small farms. Usnic acid (UA), a well-known traditional lichen metabolite, has a promising and selective antitumoral activity. However, the effects of UA on MM cells with different genomic profiles in the BRAF aspect have not been investigated yet. In this study, we evaluated the effect of UA on BRAFV600E mutated-A2058 and wild-type MeWo cells.Materials and Methods: In the UA-treated cells, viability and cell death analysis were performed by using WST-1 and Annexin-V assays. Then, the death-related morphological changes were visualized by acridine orange(AO)/ethidium bromide (EB) staining. The cell cycle regulatory effect of UA was determined. Finally, time-dependent detection of acidic vesicular organelles (AVOs) was performed by live-cell imaging.Results: While MeWo viability significantly reduced to 53.8% and 28.6%, A2058 viability was detected as 61.3% and 50.3% at 50 and 100 mu M UA for 48 h. Thus, MeWo cells were found to be more sensitive to UA. Annexin-V and morphological analysis results showed that UA triggered mainly a vacuole-dependent cell death by the formation of AVOs, instead of apoptosis, in the MM cells. This effect was prominent in A2058 compared to MeWo. UA also slightly triggered apoptosis in MeWo cells. This study aimed to evaluate the frequency of clinically overt or silent CD in patients diagnosed with autoimmune endocrinopathy and the clinical effects of silent CD in these endocrinopathies. The study included 166 patients with known or newly diagnosed mono-/polyglandular AED and 90 age- and gender-matched healthy controls. The patients were classified into four groups: type 1 diabetes mellitus (DM) (n=44), Hashimoto's thyroiditis (HT) (n=68), Addison's disease (AD) (n=17), and autoimmune polyglandular syndrome (APS) (n=37). All subjects were serologically screened for tissue transglutaminase antibody (tTG) IgA and IgG. In addition, to evaluate the possible systemic consequences of CD, serum parathormone (PTH), 25-hydroxicholecalsiferol (25-OH-Vit D), vitamin B12, folic acid, iron, iron-binding capacity (IBC), and ferritin levels were measured. In the total series, 193 (75.4%) individuals were females, and 63 (24.6%) were males. TTG IgA antibody positivity was found in 23 among 166 patients, while no positivity was encountered in the healthy control group. The highest rates of positive tTg IgA frequency were detected in AD, with 29.4% (5/17). Serum 25-OH-Vit D, vitamin B12, folic acid, iron, and ferritin levels were significantly lower in AEDs compared to controls (p<0.001), and the lowest these parameters were detected in patients with AD. The serologic CD prevalence is higher in autoimmune mono-/and polyglandular endocrine diseases than in the control group. Through the use of an alternative promoter, a truncated c-KIT protein (tr-KIT) of 30-50 kDa is generated, lacking the extracellular and transmembrane domain. Tr-KIT promotes the formation of a multi-molecular complex composed of Fyn, Plc gamma 1, and Sam68. Imatinib blocks the activity of full-length c-KIT but has no effect on tr-KIT. LNCaP is the human PCa cell line that shows tr-KIT overexpression and PC3 does not show tr-KIT overexpression. miR-128/193a-5p/494 are miRNAs targeting FYN, PLC gamma 1, and SAM68 combinatorially. The study's question is: can miR-128/193a-5p/494 be related to imatinib resistance in PCa?Methods: LNCaP and PC3 cells were treated with imatinib in IC50 doses. Before and after imatinib administration, RNA was isolated and cDNA conversion was performed. By qPCR analysis, expression changes of tr-KIT specific pathway elements and miR-128/193a-5p/494 were analyzed before and after imatinib administration.Results: After imatinib administration, miR-128/193a-5p/494 were significantly overexpressed in LNCaP cells while downregulated significantly in PC3 cells (p<0.05). Also, FYN was upregulated in LNCaP cells (p<0.05) but there was no change in PC3 after imatinib administration.Conclusion: Especially upregulation of FYN may sponge miR128/193a-5p/494 and downregulate their transcriptional activity in LNCaP cells having tr-KIT activity. Records of 143 patients (108 F, 35 M) were reviewed in a multicenter study. At diagnosis., 38% of patients were prepubertal. Anti-thyroid drugs (ATD) were used as initial therapy. There was no significant difference in clinical and laboratory characteristics at diagnosis, during treatment and adverse reaction to ATD with respect to pubertal status. Twenty patients (7 prepubertal, 13 pubertal) reached remission on ATD. Surgery was performed in seven and radioiodine (RAI) in four patients. Duration of treatment needed to achieve remission was longer in prepubertal (4.2 +/- 1.0 yr) than in pubertal patients (3.1 +/- 1.3 yr) (p = 0.02). The rate of remission was not different between prepubertal (25.9%) and pubertal patients (33.3%) (p = 0.59). ATD were associated with low remission rate in pediatric GD and required longer duration of therapy in prepubertal patients. We aimed to investigate their validity and usefulness by reevaluating the electroencephalogram (EEG) examinations of our patients in this study. Materials and Methods: We retrospectively evaluated all EEG recordings of patients diagnosed with NCSE by experienced clinical neurophysiologists in our EEG laboratory over a period of 2 years. Two neurologists trained in EEG reanalyzed all EEG data and categorized these patients as NCSE, possible NCSE, or non-NCSE using the SCC-NCSE. Results: Twenty-nine patients with a mean age of 31.5 & PLUSMN; 25.9 were reanalyzed. According to the SCC-NCSE, 24 patients (82.7%) were diagnosed as NCSE. Eighteen patients (62%) who fulfilled all SCC-NCSE were diagnosed as NCSE, whereas six patients (20.7%) were diagnosed only as possible NCSE. Five patients (17.3%) did not fulfill SCC-NCSE; the reasons are the lack of additional secondary criteria in 2 patients with encephalopathy, the absence of full compliance with the criteria in other 2 patients, and a diagnosis of electrical status epilepticus during sleep in the last patient. Conclusion: The results of our study show that SCC-NCSE is highly consistent with clinical practice to decide for the diagnosis of NCSE. The evaluation of NCSE according to a set of new standardized criteria is thought to be difficult in practice, but it provides a more objective assessment. PCR, a molecular diagnostic method, was used for the identification of the parasite species. Microscopic examination revealed that out of the 45 ovine cardiac tissue samples examined 23 contained Sarcocystis spp. cysts. The PCR analysis results showed that out of the 23 cardiac tissue specimens infected with sarcocysts, 7 were determined to be infected with Sarcocystis gigantea and 16 with Sarcocystis tenella. The Yang's local fractional calculus tool has fulfilled a significant character in defining the fractal behaviours in a fractal space or microgravity space that arise in applied nonlinear sciences. The travelling wave transformation of the non-differentiable type is introduced and we retrieve successfully the non-differentiable exact traveling wave solutions (soliton pulses in (2+1)-dimensions) of LFKMN equation with aid of generalized exp-function method in the form of generalized functions described on Cantor sets. With the help of Mathematica package program, 3D graphs were drawn for the special values of the parameters in the solutions, and the physical structures of the solutions obtained in this way were also observed. The solutions obtained can be used in the explanation of physical phenomena occurring in propagation of rogue waves in oceans and higher order optical solitons in optical fibers in current-like nonlinearities.The deduced explicit solutions will cause a new pathway of the nonlinear wave theory by the help of local fractional derivative. We investigated the correlation between percutaneous biliary drainage technique and infection frequency by evaluating patients with clinical findings, bile and blood cultures, complete blood counts, and blood biochemistry.Results: There was no statistically significant difference between the selected patient groups (internal-external or external biliary drainage catheter placed) in terms of age, sex, primary diagnosis, receiving chemotherapy, catheter sizes, and outpatient-patient status. After catheterization, catheter-related infection was observed in 49 of 216 (22.7%) patients with internal-external and 18 of 127 (14.2%) patients with external biliary drainage catheters, according to the defined criteria. There was no difference in infection rate after the biliary drainage in the two groups (P > 0.05). Usnic acid (UA) is a dibenzofuran derivative isolated from lichens. This study aimed to investigate the effects of usnic acid on inhibition of ocular biofilm formation due to CoNS.Material/Methods: Nine Staphylococcus epidermidis isolates, 5 Staphylococcus hominis isolates, 2 Staphylococcus saprophyticus iso-lates, and 1 Staphylococcus capitis and Staphylococcus lentus isolates were taken as test bacteria. They were inoculated into brain heart infusion broth and incubated for 24 hours at 35 degrees C and activated. Antibiotic suscep-tibility was investigated by Kirby-Bauer disc diffusion method. Biofilm production was determined using the microtiter plate method and optical densitometry was measured at 570 nm using an automated microplate reader. Anti-biofilm activity of UA was determined by microtitration method and biofilm removal percentage was calculated.Results: All tested bacteria were found as high biofilm-producer strains; they were generally resistant to methicillin, but susceptible to vancomycin. UA inhibited the biofilm formation of S. epidermidis isolates, ranging from 5.7% to 81.5%. It inhibited the biofilm formation of S. saprophyticus and S. lentus by 73.3% and 74.3%, respective-ly. There was no effect of UA on mature biofilms of S. epidermidis 17.7H, S. epidermidis 15.41, S. hominis 9.3, S. hominis 17.2H, S. saprophyticus, and S. lentus.Conclusions: It was determined that UA exerted anti-biofilm activity on some CoNS isolated from the ocular surface. Firstly, the nanobeam has been modeled by placing two springs that can be deformed in the downward direction. These springs control the amount of downward displacement at the ends. In the analytical solution, the displacement points are defined by two coefficients and the interior part of the nanobeam deflection is expressed by Fourier sine series. Stokes' transformation is preferred to enforce the boundary conditions to the desired point. After the mathematical operations, a matrix of coefficients including the general elastic spring constants has been found. The eigenvalues of this coefficient matrix give the frequencies of the Levinson nanobeam. The effect of some parameters on the free vibration frequencies is shown in a series of graphs and tables.
