Abstract
Dünya genelinde ve ülkemizde insanlarda ve hayvanlarda en yaygın ve en önemli kanser türlerinden biri olan meme kanserleri maddi ve manevi olarak ciddi kayıplara yol açmaktadır. Hastalığın erken teşhis edilememesine ve tedavi süreçlerinin zorlu, pahalı ve kesin sonuç vermemesine bağlı olarak prognostik faktörler ve erken teşhis oldukça önemli bir hal almaktadır. Bağışıklık sisteminin en önemli hücrelerinden biri olarak gösterilen mast hücreleri kanser dokularında önemli derecede arttıkları bilinse de olası rolleri ve fenotipleri henüz kesin olarak bilinmemektedir. Tez çalışmasında deneysel yolla sıçanlarda oluşturulan meme karsinomu ve kedi, köpek doğal olarak meydana gelen meme tümörü olgularında mast hücrelerinin bu bölgedeki olası yerleşim yerleri, varlıklarının tespiti, olası rolleri ve immunofenotipik özellikleri incelenmiştir. Çalışmada ayrıca TNF-alfa, MMP-9 ve PCNA ekspresyonları incelenerek mast hücreleri ile olası ilişkileri sorgulanmıştır. Deneysel olarak oluşturulan sıçan ve kedi köpek meme tümörü olgularında mast hücrelerinin sayısı normal meme dokusuna göre artış gösterdiği ve özellikle intratümöral alanda çok fazla arttıkları, PCNA ekspresyonu ile negatif korelasyon göstererek mast hücrelerinin bölgede artmasının muhtemelen canlı için iyi yönde bir etki gösterdiğini düşündürmüştür. Bu bulgular ışığında mast hücrelerinin tümör dokusunda önemli roller üstlenebileceği, prognostik açıdan yararlı olabileceği sonucuna varılmıştır.
Breast cancer, which is one of the most common and important types of cancer in humans and animals worldwide and in our country, causes material and spiritual severe losses. Prognostic factors and early diagnosis become crucial since the disease cannot be diagnosed early, and the treatment processes are difficult, expensive, and inconclusive. Although mast cells are considered one of the most important cells of the immune system and are known to increase significantly in cancer tissues, their possible roles and phenotypes are not yet precise. In the thesis study, the possible localization, presence, possible roles, and immunophenotypic characteristics of mast cells in this region were investigated experimentally for both conditions; mammary tumors in rats and naturally occurring mammary carcinomas in cats and dogs. In the study, TNF-alpha, MMP-9, and PCNA expressions were also examined, and their possible relations with mast cells were questioned. The number of mast cells increased in experimental rat and cat dog mammary carcinomas cases compared to healthy mammary tissues, and the increase was drastic especially in the intratumoral area; having a negative correlation with PCNA expression suggested that the increasing number of mast cells in the region probably showed a good effect on the living being. In light of these findings, it was concluded that mast cells can play an essential role in tumor tissue and can be beneficial in terms of prognostic.
