Kanatlılardan izole edilen salmonella suşlarının identifikasyonunda kullanılan metotlar üzerinde araştırmalar

Abstract
Bu çalışmada, kanatlı hayvanlardaki (tavuk, civciv, güvercin, bıldırcın, hindi) salmonella mikroorganizmalarının durumu incelenmiş, izole edilen salmonellaların çabuk teşhisi amacı ile konvansiyonel ve çabuk mikrobiyokimyasal testlerle karşılaştırmalı olarak identifikasyonu yapılmıştır. Ekim 1993-Eylül 1994 tarihleri arasında toplam 29 kaynakta, hastalık olgusu incelenmiş, olguların 21 tanesi ticari yumurtacılarda, 9, tanesi broilerlerde, 1 tanesi, kuluçkahanede, 1 tanesi yumurtacı damızlıkta, 1 tanesi kanatlı mezbahasında, 1 tanesi hindide, 1 tanesi güvercinde, 1 tanesi bıldırcında saptanmıştır. 29 kaynaktan izole edilen salmonellaların serotiplendirme sonuçlarına göre 16 adetinin (%55,2) S.gallinarum, 9 adetinin (%31 ) S. enterit idi s, 4 adetinin (13,8) S. typhimurium olduğu tesbit edilmiştir. İzole edilen salmonellalar, çabuk mikrobiyokimyasal ve konvansiyonel testlerle karşılaştırmalı olarak identifiye edilmiştir. Toplam 15fi şüpheli koloni muayene edilmiş, lizin dekarhoksi1az testinde 3 yanlış negatif sonuç vermiş olup uygunluk yüzdesi %9R, ürede 1 yanlış pozitif sonuç, vermiş olup uygunluk yüzdesi %99, indolde hiç yanlış sonuç vermemiş olup uygunluk yüzdesi %10O, malonatta 3 yanlış pozitif sonuç, vermiş olup uygunluk yüzdesi %9R olarak bulunmuştur. Antihiyogram test. sonuçlarına göre en fazla duyarlılık, flouroquinolon grubu ilaçlara, en fazla dirençlilik ise, eritromisine karşı bulunmuştur.Çalışma sonunda salmonellaların, araştırmada incelenen tüm kanatlılarda görüldüğü tesbit edilmiş, çabuk mirobiyokimyasal testlerin %98,R uygunluk oranı ile salmonellaların çabuk teşhisinde yararlanılabileceği gözlenmiştir.
In this study, salmonella spp. were investigated in different, species of birds, (chicken, pigeon, quail and turkey) and identification of salmonella strains were made by conventional ana rapid mi crobiochemical test systems. Both systems were compared. 29 salmonella cases were investigated between October 1993 and September 1994. Out of 29 cases, 21 of them in commercial layers, 2 in broilers, 1 in hatchery, 1 in layer breeder, 1 in broiler slaughter house, 1 in turkey, 1 in pigeon, 1 in quail were observed. The isolated strains from 29 cases were sero typed and found that Ifi (55,2%) of them S.gallinarum, 9 (31%) of them S.enteritidi s and 4 (13,8%) of them S. typhimurium. The isolates were identified by both rapid microbiochemical and conventional tests and the results were compared. Total 156 suspected colonies were investigated. The agreements between results were 98% in lysine deoarhxylase test with three negative results, 99% in urea test with one false positive result, 100% in indole test with no false results 98% in malonat test with three negative results. Antibiotic sensitivity tests were also performed and most of the isolated were found to be sensitive to f louroquinolones while most of them were resistant against erithromycine. It was concluded that salmonellosis can be seen in any type of poultry in this study. The rapid microbiochemical tests can 3effectively be used for diagnosis of salmonellosis with the agreement of 98,8%.
