Abstract
GİRİŞ: Bu çalışmanın amacı benign prostat hiperplazisi (BPH), prostatik intraepitelyal neoplazi (PİN), düşük ve yüksek grade prostat adenokarsinomalı bir grup hastada proliferatif aktivitenin bir göstergesi olan Kl-67 (MIB-1) antijeni varlığının araştırılıp, bu lezyonlardaki Kİ-67 proliferatif aktivite indeksinin kıyaslanmasıdır. MATERYAL VE METOD: Bu çalışmada Uludağ Üniversitesi Tıp Fakültesi Patoloji Anabilim Dalı'nda, Ocak 1995-Aralık 2000 yılları arasında tanı almış, 25 benign prostat hiperplazisi, 14 prostatik intraepitelyal neoplazi, 17 düşük grade ve 25 yüksek grade prostat adenokarsinoma olgusu incelendi. Bu olguların parafin bloklarından hazırlanan kesitlere immünohistokimyasal olarak Kİ-67 antikoru uygulandı. Bu dört grup lezyonun ortalama Kİ-67 proliferatif aktivite indeksleri bulunup, birbirleriyle kıyaslandı. BULGULAR: Ortalama Kİ-67 proliferasyon indeksi (%o); BPH'ta 24.12 ± 12.59, PİN'de 26 ± 28.44, düşük grade adenokarsinomada 32.17 ± 26.08 ve yüksek grade adenokarsinomada 122.92 ±84.17 olarak belirlendi. BPH, PİN ve düşük grade adenokarsinomaya ait proliferasyon indeksleri arasında anlamlı bir fark bulunmazken, yüksek grade adenokarsinomaya ait proliferasyon indeksinde, diğer üç lezyona göre anlamlı bir fark bulundu. SONUÇ: Kİ-67 proliferasyon indeksinin tümör grade'ini göstermede ve tümöral proliferatif lezyonlan açıklamada yardımcı bir yöntem olarak kullanılabileceği düşünülmüştür.

MATERIAL and METHOD: In this study, we have examined 25 benign prostatic hyperplasia, 14 prostatic intraepithelial neoplasia, 17 low and 25 high grade prostatic adenocarcinomas that were diagnosed between January-1995 and December-2000 in Uludag University Faculty of Medicine, Departmant of Pathology. The sections of paraffin blocks of these lesions were treat with Ki-67 antibody immunohistochemically. Mean Ki-67 proliferative activity indexes of these 4 group lesions were calculated and compared. RESULTS: The mean Ki-67 proliferation index (%o) was determined as; 24.12 ± 12.59 in BPH, 26 ± 28.44 in PIN, 32.17 ± 26.08 in low grade adenocarcinoma and 122.92 ± 84.17 in high grade adenocarcinoma. While there was no significant difference between BPH, PIN and low grade adenocarcinoma proliferation indexes, there found significant difference between high grade adenocarcinoma and other 3 lesions. CONCLUSION: Ki-67 proliferation index can be used as a tool to determine tumor grade and to explain tumoral proliferative lesions.
